Mechanistic insights into an atypical interaction between ATG8 and SH3P2 in Arabidopsis thaliana

Shuangli Sun et al.

Summary: The study uncovered an atypical interaction mode of Arabidopsis ATG8f with the plant unique adaptor protein SH3P2, showing unique conformational changes in ATG8f upon binding to the AIM sequence of the autophagic receptor NBR1. Additionally, the gel filtration assay revealed that the ubiquitin-associated domain of NBR1 outcompetes the SH3 domain of SH3P2 for ATG8f interaction. Biochemical and cellular analysis demonstrated distinct interfaces used by ATG8f to interact with NBR1 and SH3P2.

Phylogenomic Insights into the Origin of Primary Plastids

Iker Irisarri et al.

Summary: This study assessed the strength of the signal for the monophyly of Archaeplastida hosts using multiple phylogenomic data sets. The results showed that the shorter individual data sets did not support host monophyly, but the combined nuclear data sets supported the monophyly of primary photosynthetic eukaryotes and suggested a potential association with plastid-lacking Picozoa.

An N-terminal conserved region in human Atg3 couples membrane curvature sensitivity to conjugase activity during autophagy

Yansheng Ye et al.

Summary: The enzyme Atg3 is crucial for autophagosome formation, facilitating the connection of LC3 to PE lipids. The N-terminal conserved region of human Atg3 communicates information from the curvature-sensing domain to the active site, promoting spatial regulation of LC3-PE conjugation on target membranes.

Beyond Autophagy: The Expanding Roles of ATG8 Proteins

Jose L. Nieto-Torres et al.

Summary: The ATG8 family proteins are key players in autophagy, participating in cargo recruitment, biogenesis, transport, and fusion with lysosomes. Recent studies have shown their involvement in diverse processes such as vesicle maturation and cargo specification, with complex regulatory mechanisms that are still not fully understood. Individual roles of different ATG8 family members in canonical or non-canonical pathways remain unclear. This review highlights the molecular questions surrounding ATG8s that are only beginning to be explored.

Summary: Researchers evaluated the chronological possibility of proposed plastid origins and transmissions in eukaryotic phytoplankton through Bayesian molecular clock analyses. The study established a timeframe for the origin of red algal-derived plastids under scenarios of serial endosymbiosis, revealing that the hypotheses of serial endosymbiosis are chronologically possible.

Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)

Daniel J. Klionsky et al.

Summary: The guidelines presented in this article aim to assist researchers in selecting and interpreting methods to examine autophagy, as well as to help reviewers provide constructive critiques of reports focused on these processes. It emphasizes that no single assay is perfect for every situation, and multiple techniques may be needed to properly monitor autophagy in different experimental settings. The article also highlights the importance of targeting multiple autophagy-related genes in genetic approaches to block autophagy effectively.
