The role of acetaldehyde dehydrogenase 2 in the pathogenesis of liver diseases

Yin-cui Wu et al.

Summary: Common liver tissue damage is caused by the accumulation of toxic aldehydes in lipid peroxidation under oxidative stress. Acetaldehyde dehydrogenase 2 (ALDH2) can effectively metabolize these toxic aldehydes, alleviating various liver diseases. This review explores the relationship between ALDH2 and liver diseases such as alcoholic liver disease (ALD), non-alcoholic fatty liver disease (NAFLD), liver fibrosis, and hepatocellular carcinoma (HCC), as well as the regulatory mechanism of ALDH2 in these diseases.

Sphingosine 1-phosphate lyase facilitates cancer progression through converting sphingolipids to glycerophospholipids

Baasanjav Uranbileg et al.

Summary: This study reveals the potential role of S1P metabolism and its products in cancer pathogenesis, especially in hepatocellular carcinoma and colorectal cancer. The overexpression of SPL is associated with elevated levels of glyceroLPLs and promotes tumor progression, while the inhibition of SPL has the opposite effect. Furthermore, SPL and glyceroLPLs play important roles in regulating cancer progression through various mechanisms.

Effect of Quercetin and Fingolimod, Alone or in Combination, on the Sphingolipid Metabolism in HepG2 Cells

Albena Momchilova et al.

Summary: This study analyzed the effects of quercetin and fingolimod on sphingolipid metabolism in HepG2 cells. The results showed that quercetin and fingolimod, alone or in combination, can reduce sphingolipid levels and activate certain enzymes. The up-regulation and down-regulation of specific enzymes and proteins by quercetin have important implications for cancer treatment.

Epidemiology of Hepatocellular Carcinoma

Katherine A. McGlynn et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), is a major global health concern. While HBV and HCV remain important risk factors for HCC, efforts in vaccination and treatment are showing promise. However, the increasing prevalence of metabolic risk factors like obesity and diabetes, as well as other factors like excessive alcohol consumption, present ongoing challenges in the prevention of liver cancer.

Overriding sorafenib resistance via blocking lipid metabolism and Ras by sphingomyelin synthase 1 inhibition in hepatocellular carcinoma

Haofeng Lu et al.

Summary: The study showed that SMS1 plays a crucial role in sorafenib resistance in HCC cells by promoting growth and migration, alleviating toxicity, and disrupting sphingolipid metabolism. Inhibition of SMS1 led to growth inhibition, apoptosis induction, and increased efficacy of sorafenib in resistant cells, suggesting a potential therapeutic strategy to overcome sorafenib resistance in HCC.

Transformed ALDH2-/- hepatocytes by ethanol could serve as a useful tool for studying alcoholic hepatocarcinogenesis

Xiu-Ning Zhang et al.

Summary: Studying the transformation of immortalized normal liver cells induced by ethanol may provide a versatile tool for researching alcoholic liver cancer. Knockout of ALDH2 could potentially shorten the transformation process and increase the accumulation of acetaldehyde in hepatocytes.

Hepatocellular carcinoma

Josep M. Llovet et al.

Summary: Liver cancer, particularly hepatocellular carcinoma (HCC), poses a significant global health challenge with rising incidence projected to exceed 1 million cases by 2025. Infection by hepatitis B and C viruses remains a major risk factor for HCC, while non-alcoholic steatohepatitis is emerging as a more common risk factor in Western countries. Advances in systemic therapies for HCC, including immunotherapies and targeted therapies, are expected to revolutionize the management of this disease.

Cell surface sphingomyelin: key role in cancer initiation, progression, and immune evasion

Hatem Tallima et al.

Summary: Cell surface biochemical changes, particularly the excessive increase in outer leaflet sphingomyelin (SM) content, play a crucial role in cancer development and growth. Surface membrane SM affects cell-cell contact inhibition, cellular signal transduction, metabolic pathways, and susceptibility to host immune cells.

Summary: This study found elevated levels of sphingolipids, including S1P and ceramide, in HCC compared to surrounding normal liver tissue, indicating the important role of S1P in HCC tumor progression, warranting further investigation.
