Purification identification and function analysis of ACE inhibitory peptide from Ulva prolifera protein

Zhiyong Li et al.

Summary: In this study, Ulva prolifera, an edible alga, was used to prepare angiotensin-I converting enzyme (ACE) inhibitory peptide. The highest ACE inhibitory activity was obtained from the hydrolysate of algae protein, which was processed using various methods and identified as the peptide DIGGL. DIGGL exhibited non-competitive inhibition against ACE and mainly achieved this effect through three conventional hydrogen bonds. It also showed the ability to activate endothelial nitric oxide synthase and decrease endothelin-1 secretion in human umbilical vein endothelial cells. Moreover, DIGGL promoted splenocyte proliferation and remained active during gastrointestinal protease digestion.

Identification and molecular mechanism of novel tyrosinase inhibitory peptides from the hydrolysate of 'Fengdan' peony (Paeonia ostii) seed meal proteins: Peptidomics and in silico analysis

Yuan Zhao et al.

Summary: In this study, a hydrolysate with high tyrosinase inhibitory activity was prepared from peony seed meal proteins using Neutrase. Molecular docking showed that 38 out of 39 peptides can potentially act as tyrosinase inhibitors with lower or the same binding energies as the standard kojic acid. Three potent peptides were identified and their interaction mechanisms were analyzed, highlighting the importance of hydrogen bonding and hydrophobic interactions. In vitro tests demonstrated that these peptides had strong tyrosinase inhibitory abilities. These findings suggest that peony seed meal proteins can be a valuable source of natural tyrosinase inhibitory peptides for various industrial applications.

Integration of Molecular Docking Analysis and Molecular Dynamics Simulations for Studying Food Proteins and Bioactive Peptides

Abraham Vidal-Limon et al.

Summary: The use of in silico tools, such as molecular docking, to study biological activity of proteins and peptides can sometimes produce results that do not correlate with actual experimental binding affinities. Molecular dynamics simulations (MDS) offer a solution to overcome sampling limitations in docking analysis and provide valuable information in deciphering functional mechanisms of proteins and peptides. This review discusses the traditional use of in silico models, like molecular docking, and highlights the importance of MDS in predicting structural and functional dynamics of food proteins and bioactive peptides.
