A Novel Ageladine A Derivative Acts as a STAT3 Inhibitor and Exhibits Potential Antitumor Effects

Na He et al.

Summary: In this study, several ageladine A derivative compounds were synthesized and compound 25 was found to be the most effective. Compound 25 showed inhibitory effects on STAT3 activation, reducing the expression of downstream genes and suppressing tumor cell proliferation and migration. In vivo experiments demonstrated that compound 25 effectively inhibited the growth of A549 tumors without significant weight loss.

Eucalyptol antagonized the apoptosis and immune dysfunction of grass carp hepatocytes induced by tetrabromobisphenol A by regulating ROS/ASK1/JNK pathway

Wenying Sun et al.

Summary: In this experimental study, the mechanism of TBBPA toxicity on Grass carp hepatocytes (L8824 cells) and the antagonistic impact of EUC on TBBPA were investigated. The results showed that TBBPA exposure induced oxidative stress and activated the ASK1/JNK signaling pathway, resulting in apoptosis. Additionally, EUC effectively protected against TBBPA-induced apoptosis and immune dysfunction by regulating the ROS/ASK1/JNK pathway.

The joint effects of nanoplastics and TBBPA on neurodevelopmental toxicity in Caenorhabditis elegans

Kunming Zhao et al.

Summary: This study investigated the neurodevelopmental toxicity induced by combined exposure of TBBPA and polystyrene NPs in C. elegans. The results showed that combined exposure had synergistic inhibitory effects on survival rate, body length/width, and locomotor ability. The overproduction of ROS, lipofuscin accumulation, and dopaminergic neuronal loss indicated the involvement of oxidative stress. The increased expressions of pink-1 and hop-1 genes played an important role in neurodevelopmental toxicity induced by TBBPA and polystyrene NPs.

Melatonin alleviates lead-induced intestinal epithelial cell pyroptosis in the common carps (Cyprinus carpio) via miR-17-5p/TXNIP axis

Zhiying Miao et al.

Summary: This study investigates the effect of melatonin on lead-induced intestinal epithelial cell pyroptosis and demonstrates that melatonin attenuates pyroptosis through the miR-17-5p/TXNIP axis. The findings suggest that melatonin could be a potential therapeutic option for lead-induced intestinal injury and inflammation.

Oxidative stress mediated by the TLR4/NOX2 signalling axis is involved in polystyrene microplastic-induced uterine fibrosis in mice?

Hao Wu et al.

Summary: Microplastics can induce uterine fibrosis in female mice by triggering oxidative stress and activating the Notch and TGF-f3 signaling pathways through the TLR4/NOX2 signaling axis. Inhibition of the TLR4/NOX2 signaling pathway can reduce the expression of fibrotic genes, suggesting it as a potential therapeutic option to alleviate the reproductive toxicity of microplastics.

Heavy metal associated health hazards: An interplay of oxidative stress and signal transduction

Jagdish Gopal Paithankar et al.

Summary: The oxidative stress induced by heavy metals and its connection with different signaling pathways are complex, which may lead to a variety of health issues. Exposure to heavy metals/metalloids such as Arsenic, Chromium, Cadmium, and Lead can trigger oxidative stress and subsequently cellular and organismal responses, involving regulation of multiple signaling pathways.

Naringenin prevents pregnancy-induced hypertension via suppression of JAK/STAT3 signalling pathway in mice

Bide Duan et al.

Summary: Naringenin significantly reduced blood pressure, total urine protein level, plasma levels of VCE, alpha-ADR, and angiotensin in PIH mice. It also decreased pro-inflammatory cytokines and oxidative stress markers while increasing anti-inflammatory cytokine levels. Naringenin inhibited JAK2/STAT3 signalling by promoting SHP-1 expression, contributing to the improvement of hypertension in pregnancy.

Influence of the co-exposure of microplastics and tetrabromobisphenol A on human gut: Simulation in vitro with human cell Caco-2 and gut microbiota

Wantang Huang et al.

Summary: This study demonstrated that both PEMPs and TBBPA have detrimental effects on Caco-2 cells, with TBBPA playing a major role and high concentrations of PEMPs also affecting cell viability. TBBPA selectively inhibits the growth of gram-positive bacteria, leading to shifts in microbiota composition that are unfavorable for gut health. The interference of PEMPs and TBBPA also alters metabolism pathways of gut microbiota, highlighting the potential threat to human health.
