Small Molecules Targeting the RNA-Binding Protein HuR Inhibit Tumor Growth in Xenografts

Summary: The RNA-binding protein Hu antigen R (HuR) plays a critical role in several diseases, including cancer, and has potential as a therapeutic target. Through screening and fragment analysis, we have identified small-molecule inhibitors of HuR. Two top inhibitors, 1c and 7c, directly bind to the RNA-binding pocket of HuR, blocking its function and inhibiting cancer cell growth and invasion. In breast cancer xenograft models, inhibitor 1c shows tumor growth inhibition as a single agent and synergistic effect with chemotherapy docetaxel. Mechanistically, 1c interferes with the HuR-TGFB/THBS1 axis.

Emerging TACnology: Heterobifunctional Small Molecule Inducers of Targeted Posttranslational Protein Modifications

RNA-binding proteins that lack canonical RNA-binding domains are rarely sequence-specific

Summary: Thousands of RNA-binding proteins (RBPs) bind to cellular mRNA, including unconventional RBPs (ucRBPs) that lack known RNA-binding domains. We investigated 492 human ucRBPs for RNA-binding and identified 23 that bind specific RNA sequences. Most of these proteins have previously been associated with RNA-related functions. We found that CCHC-zf domains from seven human proteins recognize specific RNA motifs, indicating their importance as a major class of RNA-binding domains.

First Small-Molecule Inhibitors Targeting the RNA-Binding Protein IGF2BP2/IMP2 for Cancer Therapy

Summary: This study validated IMP2 as a potential target for colorectal cancer and established a screening assay for small-molecule inhibitors of IMP2. In vitro and in vivo studies demonstrated that IMP2 knockout reduces tumor cell proliferation and growth. Ten compounds from different libraries showed target specificity and were validated in vitro. The three most active compounds also exhibited reduced tumor xenograft growth in vivo. These findings support IMP2 as a druggable target to reduce tumor cell proliferation.

Our study identified 57 RBP candidates with distinct functions in supporting MYC-driven oncogenic pathways, highlighting the therapeutic potential of RBPs, especially YTHDF2, in counteracting global mRNA synthesis in MYC-driven breast cancers.
