ADP-ribosyltransferases, an update on function and nomenclature

Bernhard Luscher et al.

Summary: ADP-ribosylation, a post-translational modification of proteins, nucleic acids, and metabolites, plays diverse roles in cellular processes such as stress responses, signaling, and transcriptional regulation. Recent advances in research have identified a wide range of cellular pathways regulated by ADP-ribosylation, highlighting the importance of understanding this mechanism in cell biology.

Captured snapshots of PARP1 in the active state reveal the mechanics of PARP1 allostery

Elise Rouleau-Turcotte et al.

Summary: PARP1 can rapidly detect DNA strand break damage and activate the production of poly(ADP-ribose) by signaling break detection to its catalytic domain. This study provides insights into the contributions of the regulatory helical domain (HD) to PARP1 allostery and the interaction with DNA damage, as well as the mechanisms of PARP1 catalytic activation and retention on DNA damage.

Progress and outlook in studying the substrate specificities of PARPs and related enzymes

Marcin J. Suskiewicz et al.

Summary: Despite years of research on ADP-ribosyltransferases from the PARP family, their substrate specificity has remained unclear. Recent breakthroughs include identifying protein serine residues, cysteine, and tyrosine as potential targets of specific PARPs. These findings shed new light on PARP-mediated catalysis and caution to expect the unexpected with potential substrates.

G-Quadruplex loops regulate PARP-1 enzymatic activation

Andrea D. Edwards et al.

Summary: G-quadruplexes are non-B form DNA structures found at regulatory regions, such as promoters of proto-oncogenes and telomeres. PARP-1 interacts with G-quadruplexes and its enzymatic activation is dependent on loop features and the presence of oxidative damage. These interactions suggest a functional relationship between PARP-1 and G-quadruplexes.

The BRCT domain of PARP1 binds intact DNA and mediates intrastrand transfer

Johannes Rudolph et al.

Summary: PARP1 is a key player in DNA damage response and a target for cancer treatment. The BRCT domain binds to DNA without activating the catalytic domain. The DNA-binding properties of the BRCT domain contribute to the monkey-bar mechanism for DNA transfer of PARP1.

Identifying Poly(ADP-ribose)-Binding Proteins with Photoaffinity-Based Proteomics

Morgan Dasovich et al.

Summary: The study synthesized a PAR photoaffinity probe to identify numerous PAR binding proteins involved in DNA repair, RNA processing, and metabolism, demonstrating that polymer length may regulate the outcome and timing of PAR signaling pathways. This investigation provides the first census of PAR-binding proteins, offers a proteomics analysis of length-selective PAR binding, and associates PAR binding with RNA metabolism and the formation of biomolecular condensates.

SPINDOC binds PARP1 to facilitate PARylation

Fen Yang et al.

Summary: SPINDOC interacts with histone code effector protein SPIN1 and forms two distinct protein complexes, one with SPIN1 and the other with PARP1. It plays a role in regulating PARP1-mediated PARylation and the DNA damage response. Knockout of SPINDOC leads to reduced PARylation levels, smaller body size in mice, and hypersensitivity to IR-induced DNA damage.

Dual function of HPF1 in the modulation of PARP1 and PARP2 activities

Tatyana A. Kurgina et al.

Summary: The histone PARylation factor 1 (HPF1) has been found to stimulate the autoPARylation of PARP1 and PARP2 as well as the heteroPARylation of histones in the complex with nucleosome. The stimulatory action is dependent on the concentrations of HPF1 and NAD(+), with PARP2 showing more efficiency than PARP1 in the reaction. The dual function of HPF1 in maintaining PARP activity is also discussed.

Serine-linked PARP1 auto-modification controls PARP inhibitor response

Evgeniia Prokhorova et al.

Summary: Recent findings suggest that serine ADP-ribosylation plays a vital role in cellular responses to PARP1/PARP2 inhibitors. The efficient modification of three serine residues within PARP1 by HPF1 counters PARP1 trapping and contributes to inhibitor tolerance, implicating these residues as potential biomarkers for PARP inhibitor therapy.
