Aging weakens Th17 cell pathogenicity and ameliorates experimental autoimmune uveitis in mice

He Li et al.

Summary: Aging-induced changes in the immune system are associated with a higher incidence of infection and vaccination failure. In old mice, immune cell responses, especially Th17 cells, are altered, leading to a milder autoimmune response compared to young mice. This study highlights the role of GM-CSF secretion and IL-23R levels in regulating autoimmune challenges in aged mice.

mRNA modifications in cardiovascular biology and disease: with a focus on m6A modification

Rajesh Kumari et al.

Summary: m(6)A methylation is a highly studied RNA modification that plays a crucial role in cellular functions during development, differentiation, and disease. It is found in different RNA species and is regulated by various enzymes and proteins. In cardiovascular diseases, m(6)A methylation is associated with cardiac hypertrophy, myocardial infarction, and atherosclerosis.

Extracellular vesicle-packaged mitochondrial disturbing miRNA exacerbates cardiac injury during acute myocardial infarction

Ping Sun et al.

Summary: Mounting evidence suggests that extracellular vesicles (EVs) play an important role in cardiac physiology and pathology. This study found that EVs induced by acute myocardial infarction (AMI) can impair cardiomyocyte survival and worsen cardiac injury. The mechanism involves the EV-encapsulated miR-503 directly targeting PGC-1β and SIRT3, leading to mitochondrial metabolic dysfunction and cardiomyocyte death. Endothelial cells were identified as the primary source of miR-503 in EVs after AMI.

MYC-mediated silencing of miR-181a-5p promotes pathogenic Th17 responses by modulating AKT3-FOXO3 signaling

Sisi Chen et al.

Summary: This study reveals that miR-181a-5p negatively regulates pathogenic Th17 cell responses by targeting AKT3 and activating FOXO3. Decreased miR-181a-5p and up-regulated AKT3 expression were found in uveitis patients. Intravitreal administration of miR-181a-5p mimics effectively attenuated clinical and pathological signs of established uveitis.

Dendritic Cell Regulation of T Helper Cells

Xiangyun Yin et al.

Summary: Dendritic cells play a crucial role in sensing the microenvironment and shaping the adaptive immune response. They consist of several subsets that regulate T lymphocyte differentiation and determine distinct CD4(+) T helper cell differentiation outcomes. The intrinsic properties of DC subsets, local tissue microenvironments, and interactions with other immune cells contribute to Th cell differentiation during homeostasis and inflammation.

Exosomal miR-338-3p suppresses non-small-cell lung cancer cells metastasis by inhibiting CHL1 through the MAPK signaling pathway

Wen Tian et al.

Summary: This study evaluated the specific regulatory mechanisms of the exosomal miR-338-3p/CHL1/MAPK signaling pathway axis in non-small-cell lung cancer. The upregulation of CHL1 enhanced tumor cell proliferation and inhibited apoptosis, while miR-338-3p was found to mediate tumor cell progression by directly interacting with CHL1. Exosomal miR-338-3p suppressed tumor cell proliferation and metastasis by inhibiting the activation of the MAPK signaling pathway through downregulating CHL1 expression.

Summary: This study aimed to characterize microRNA-related regulatory mechanisms in the aorta during atherosclerosis. The research analyzed miRNA expression changes in different cell types under proatherogenic stimuli and found common miRNA species dominating in all cell types. The study identified potential mechanisms by which miRNAs affect atherogenesis in a cell-type-specific manner.

Activation of Transcription Factor 4 in Dendritic Cells Controls Th1/Th17 Responses and Autoimmune Neuroinflammation

Indumathi Manoharan et al.

Summary: The activation of the TCF4 transcription factor in dendritic cells is critical for controlling the magnitude of inflammatory responses and limiting neuroinflammation. Loss of TCF4 in DCs leads to heightened activation of p38 MAPK and increased levels of proinflammatory cytokines, exacerbating experimental autoimmune encephalomyelitis pathology. Manipulation of the TCF4 pathway in dendritic cells represents a potential therapeutic approach to control autoimmune neuroinflammation.
