An Original Aspirin-Containing Carbonic Anhydrase 9 Inhibitor Overcomes Hypoxia-Induced Drug Resistance to Enhance the Efficacy of Myocardial Protection

Wen Zhou et al.

Summary: The overexpression of CAIX in myocardial cells is closely related to drug resistance in myocardial ischemia injury. A novel CAIX inhibitor, AcAs, has been developed to overcome acquired drug resistance and more effectively attenuate myocardial ischemia and hypoxia injury than aspirin. CAIX inhibitor shows great potential for further application in myocardial hypoxia injury therapy.

A hypoxia-activated NO donor for the treatment of myocardial hypoxia injury

Wen Zhou et al.

Summary: In this study, a novel hypoxia-activated NO donor (Hano) was designed and prepared, which can increase NO release, dilate blood vessels, and attenuate myocardial hypoxia injury. This drug shows great potential for the treatment of hypoxic heart diseases.

Inflammation and cardiovascular diseases: lessons from seminal clinical trials

Luca Liberale et al.

Summary: Canakinumab Anti-inflammatory Thrombosis Outcome Study (CANTOS) and Colchicine Cardiovascular Outcomes Trial (COLCOT) were the first two randomized clinical trials to demonstrate the effectiveness of specific anti-inflammatory treatments in cardiovascular prevention, confirming the involvement of specific inflammatory pathways in atherosclerotic cardiovascular disease. Other trials targeting multiple inflammasome-independent pathways failed to show benefit, highlighting the significance of balancing the risk of impaired host defense and infections with the prevention of major adverse cardiovascular events in patients with residual inflammatory risk.

Emerging role of carbonic anhydrase inhibitors

Claudiu T. Supuran

Summary: Inhibition of carbonic anhydrase has been used clinically for various applications including treating glaucoma, epilepsy, obesity, and cancer. Recent studies have discovered new roles of CA isoforms in areas like pain management, Alzheimer's disease, and oxidative stress, leading to potential novel clinical applications. Additionally, inhibition of CA in pathogenic microorganisms is being explored for new anti-infective drugs.
