Impact of the PEG length and PEGylation site on the structural, thermodynamic, thermal, and proteolytic stability of mono-PEGylated alpha-1 antitrypsin

Xiao Liu et al.

Summary: Conjugation to polyethylene glycol (PEG) is a commonly used method to improve the therapeutic value of proteins. This study investigated the effects of PEGylation on the structure, stability, and polymerization of alpha1-antitrypsin (AAT). The results showed that PEGylation did not significantly change the structure or stability of AAT, but decreased its propensity to aggregate upon heat treatment and improved its proteolytic resistance.

Biodistribution and elimination pathways of PEGylated recombinant human deoxyribonuclease I after pulmonary delivery in mice

Sohaib Mahri et al.

Summary: The conjugation of rhDNase with PEG30 kDa significantly extends its residence time in mouse lungs, ideal for its mucolytic action. PEGylated rhDNase is mainly cleared through the intratracheal route, while the unconjugated form is primarily eliminated within 24 hours.

PEGylation of recombinant human deoxyribonuclease I decreases its transport across lung epithelial cells and uptake by macrophages

Sohaib Mahri et al.

Summary: Conjugation with high molecular weight polyethylene glycol (PEG) significantly prolongs the lung residence time of recombinant human deoxyribonuclease I (rhDNase) by reducing transport across lung epithelial cells and uptake by macrophages, thus improving therapeutic efficacy after pulmonary delivery.

Protein Engineering Strategies for Improved Pharmacokinetics

Aurelie Rondon et al.

Summary: Protein therapeutics have become an important pillar in treating various diseases, but their clinical application is often hindered by issues such as short half-life and immunogenicity. Strategies to extend the half-life of proteins are being developed through polymer chemistry, protein engineering, and fusion to other proteins, with a focus on balancing between half-life extension and maintaining therapeutic activity.
