Differential effects on human cytochromes P450 by CRISPR/Cas9-induced genetic knockout of cytochrome P450 reductase and cytochrome b5 in HepaRG cells

Tamara Heintze et al.

Summary: The use of lentiviral transduction to deliver Cas9 and sgRNAs in HepaRG cells resulted in efficient knockdown of POR and revealed differential effects on drug-metabolizing CYP enzymes. Knockdown of POR also affected the expression of CYPs at mRNA and protein levels. The study highlights the importance of POR/NADPH- and CYB5/NADH-electron transport systems in influencing human drug metabolism, with implications for future research on hepatic functions.

How circadian variability of the heart rate and plasma electrolytes concentration influence the cardiac electrophysiology - model-based case study

Barbara Wisniowska et al.

Summary: The circadian rhythm of cardiac electrophysiology is influenced by various physiological and biochemical factors. Modeling and simulation studies can provide insights into heart chronotherapy questions. The study evaluated the performance of circadian models in Cardiac Safety Simulator v2.2 and found that electrolyte balance variability is the main factor influencing QT circadian pattern.

The Role of Epoxyeicosatrienoic Acids in Cardiac Remodeling

Jinsheng Lai et al.

Summary: Epoxyeicosatrienoic acids (EETs) are metabolites of arachidonic acid produced by cytochrome P450 (CYP) epoxygenases, with beneficial effects against inflammation, fibrosis, and apoptosis in cardiovascular diseases. Studies have shown that increasing EET levels can protect against cardiovascular diseases such as hypertension, myocardial infarction, and heart failure. Therefore, upregulation of EETs may be a potential therapeutic strategy for cardiovascular diseases.

Atypical kinetics of cytochrome P450 2J2: Epoxidation of arachidonic acid and reversible inhibition by xenobiotic inhibitors

Jacqueline Wen Hui Leow et al.

Summary: The study characterized the atypical substrate inhibition kinetics of CYP2J2 metabolism of AA to 14,15-EET, revealing complete substrate inhibition phenomenon. Molecular dynamics simulations confirmed the conformational stability of CYP2J2 with dual AA substrate binding. Eadie-Hofstee plots demonstrated that complete substrate inhibition of CYP2J2 by AA was not attenuated even at high concentrations of xenobiotic inhibitors.
