Total Synthesis of Paclitaxel

Summary: The total synthesis of paclitaxel is achieved through a double Rubottom oxidation of the bis(silyl enol ether) derived from a tricarbocyclic diketone, efficiently installing a bridgehead olefin and C-5/C-13 hydroxy groups in one step. Additionally, a novel Ag-promoted oxetane formation method is used to smoothly construct the tetracyclic framework of paclitaxel.

Bioproduction of Enantiopure (R)- and (S)-2-Phenylglycinols from Styrenes and Renewable Feedstocks

Summary: Enzyme cascades were developed to convert styrene into enantiopure (R)- and (S)-2-phenylglycinols, as well as other substituted styrenes and indene into substituted phenylglycinols and 1-amino-2-indanol with high enantioselectivity. Enantioselective enzyme cascades were also developed to produce (R)- and (S)-2-phenylglycinols from bio-based L-phenylalanine. Combining L-phenylalanine biosynthesis pathway with enantioselective cascade in one strain or coupled strains enabled the synthesis of (R)- and (S)-2-phenylglycinols from renewable feedstocks, enhancing production from glucose and glycerol.

Summary: Unfavorable equilibrium positions are crucial in the development of biocatalytic reactions, especially in transaminase-catalyzed reactions for the synthesis of enantiomerically pure amines. This study presents a process development integrating in situ-product crystallization, allowing downstream processing via filtration and required equilibrium shift. The approach demonstrates a semi-continuous reaction concept with intermediate substrate addition and product removal supported by light vacuum for by-product acetone removal, enabling higher yield at larger scales.
