Identification of a new and diverse set of Mycobacterium tuberculosis uracil-DNA glycosylase (MtUng) inhibitors using structure-based virtual screening: Experimental validation and molecular dynamics studies

Prateek Raj et al.

Summary: This study conducted the first structure-based virtual screening and discovered a series of potential compounds inhibiting Mycobacterium tuberculosis uracil-DNA glycosylase. One compound exhibited better inhibitory effect than known inhibitors. These compounds represent a diverse set of scaffolds for developing next-generation MtUng inhibitors.

A New Class of Uracil-DNA Glycosylase Inhibitors Active against Human and Vaccinia Virus Enzyme

Inga R. Grin et al.

Summary: Uracil-DNA glycosylases are crucial enzymes for the efficiency of antimetabolite drugs and virulence in certain infections. Existing UNG inhibitors are based on uracil base, but new pharmacophores like PyO3 are needed for a wider range of UNGs. PyO3 shows potential as a novel pharmacophore for UNG-targeting agents.
