Computational methods to analyze and predict the binding mode of inhibitors targeting both human and mushroom tyrosinase

Resorcinol-based hemiindigoid derivatives as human tyrosinase inhibitors and melanogenesis suppressors in human melanoma cells

Summary: Considering the crucial role of human tyrosinase (hsTYR) in melanogenesis, finding new hsTYR inhibitors is essential for targeted pharmacological and dermocosmetic applications. Despite the abundance of mushroom tyrosinase (abTYR) inhibitors, the structural differences between these enzymes necessitate the development of true hsTYR inhibitors. In this study, researchers synthesized 38 derivatives and found two compounds that showed excellent inhibition values in hsTYR assays and demonstrated promising improvement in whole cell experiments.

Evaluation of 2,3-Dihydro-1,5-benzothiazepine Derivatives as Potential Tyrosinase Inhibitors: In Vitro and In Silico Studies

Summary: Benzothiazepines were investigated for their inhibitory potential against mushroom tyrosinase. The synthesized derivatives showed moderate to good IC50 values and exhibited better inhibition compared to the reference kojic acid. Compound 2 was identified as a mixed-type tyrosinase inhibitor. Molecular modeling studies and molecular dynamic simulations supported the binding modes and potential of the analogues as antityrosinase agents.

The newly synthesized compounds showed strong inhibitory activity against tyrosinase, with compound 2n exhibiting the lowest IC50 value. It could be recommended as a potential lead candidate for treating tyrosinase-mediated hyperpigmentation in the future.
