Half-sandwich ruthenium(II), rhodium(III) and iridium(III) fluorescent metal complexes containing pyrazoline based ligands: DNA binding, and antibacterial activities

Summary: A series of new complexes of ruthenium(II), rhodium(III), and iridium(III) with pyrazoline-based ligands were synthesized and characterized. These compounds exhibited good antibacterial activity against Gram-positive bacteria and moderate antioxidant activity. They also showed moderate affinity for binding to Salmon Milt DNA and demonstrated higher cytotoxicity in Dalton's lymphoma cell lines.

Half-Sandwich Ruthenium Arene Complexes Bearing Clinically Approved Drugs as Ligands: The Importance of Metal-Drug Synergism in Metallodrug Design

Summary: A novel strategy in metallodrug discovery is incorporating clinically approved drugs into metal complexes as ligands. This approach has been used to re-purpose drugs, prepare organometallic complexes, overcome drug resistance, and design promising alternatives to current metal-based drugs. The combination of organoruthenium moiety and clinical drug in a single molecule has been shown to enhance pharmacological activity and reduce toxicity. This review summarizes recent reports on rationally designed ruthenium complexes containing FDA-approved drugs, focusing on coordination mode, kinetics, mechanism, and structure-activity relationship. It provides insights for future developments in ruthenium-based metallopharmaceuticals.

Towards Selective Delivery of a Ruthenium(II) Polypyridyl Complex-Containing Bombesin Conjugate into Cancer Cells

Summary: An increasing number of novel Ru(II) polypyridyl complexes have been successfully applied as photosensitizers (PSs) for photodynamic therapy (PDT). Classical maleimide and versatile NHS-activated acrylamide strategies were employed to conjugate a promising Ru(II) polypyridyl complex to the N-terminally Cys-modified Bombesin (BBN) targeting unit. Although lacking in tumoral selectivity, the new bioconjugates showed high phototoxic activity and decreased toxicity in the absence of light irradiation, making them potentially useful for advanced cancer treatment.

Nanoencapsulation of Ru(p-cymene) Complex Bearing Ginger-based Natural Product into Liposomal Nanoformulation to Improve Its Cellular Uptake and Antiproliferative Activity

Summary: Organometallic compounds are promising candidates in the development of metallochemotherapeutics. In this study, two Ru(II)-pcymene complexes containing natural products were synthesized and characterized. These complexes exhibited higher cytotoxicity and antioxidant activity, and were shown to accumulate in melanoma cancer cells. Their cytotoxicity was positively correlated with cell uptake and mainly resulted from the induction of singlet oxygen production in cancer cells. The complexes also moderately altered the adhesion properties of cancer cells, which is beneficial for antimetastatic treatment.

Ruthenium emerged as a promising candidate due to its significant anticancer properties and low side effects, with several ruthenium complexes entering clinical trials. Few arene ruthenium complexes have shown anti-metastatic properties in clinical trials.
