Zn(II)-Catalyzed Selective N-Alkylation of Amines with Alcohols Using Redox Noninnocent Azo-Aromatic Ligand as Electron and Hydrogen Reservoir

Summary: We developed a sustainable and eco-friendly method for selective N-alkylation of various amines using a well-defined Zn(II)-catalyst with a tridentate arylazo scaffold. A total of 57 N-alkylated amines were synthesized with good to excellent yields, including 17 new examples. The Zn(II)-catalyst demonstrated wide functional group tolerance and compatibility for the synthesis of dialkylated amines, providing high yields for the precursors of tripelennamine and thonzonium bromide drugs.

Ruthenium-Catalyzed Direct Reductive Amination of Carbonyl Compounds for the Synthesis of Amines: An Overview

Summary: Reductive amination is a valuable method for amine synthesis, which has been extensively studied in both academia and industry for a century. Amines and their derivatives are versatile building blocks for various organic substrates and play a significant role in the production of advanced chemicals, pharmaceuticals, polymers, and more. The development of innovative catalytic processes for the selective and sustainable synthesis of amines from readily available and environmentally friendly reagents remains a major focus in chemical research. Ruthenium catalysts have shown excellent performance in reductive amination due to their stability, selectivity, versatility, low toxicity, and high efficiency. This review provides a comprehensive overview of Ru-catalyzed reductive amination processes, covering the literature from 2009 to 2022.

Zn(II)-Catalyzed Multicomponent Sustainable Synthesis of Pyridines in Air

Summary: In this study, a Zn(II)-catalyzed solvent-free sustainable synthesis of tri- and tetra-substituted pyridines was reported. The primary feedstock used was alcohols and NH4OAc served as the nitrogen source. The well-defined air-stable Zn(II)-catalyst, featuring a redox-active tridentate azo-aromatic pincer, was used for the synthesis of a wide variety of unsymmetrical 2,4,6-substituted pyridines by three-component coupling of alcohols with NH4OAc. The reactions proceeded efficiently and produced high yields of the desired products.

N-Alkylation of Amines by C1-C10 Aliphatic Alcohols Using A Well- Defined Ru(II)-Catalyst. A Metal-Ligand Cooperative Approach

Summary: A Ru(II)-catalyzed N-alkylation reaction of amines with C1-C10 aliphatic alcohols was described. The air-stable catalyst 1a exhibited wide functional group tolerance and high catalytic efficiency, requiring only low catalyst loading. Various N-methylated, N-ethylated, and N-alkylated amines were efficiently prepared. The reaction proceeded through a borrowing hydrogen transfer pathway and showed excellent chemo-selectivity.

N-Alkylation of organonitrogen compounds catalyzed by methylene-linked bis-NHC half-sandwich ruthenium complexes

This is the first example of N-alkylation of sulfonamides using transition-metal complexes with NHC ligands.

Evolving New Chemistry: Biocatalysis for the Synthesis of Amine-Containing Pharmaceuticals

It has found application in the development of small molecule pharmaceuticals, particularly for amine-containing drugs. This review discusses the use of biocatalysis in creating safer, more efficient, and cheaper processes for the manufacture of chiral amine-containing pharmaceuticals and alkaloids. Different enzyme classes have been utilized, and the potential of biocatalysis in synthesizing natural products and high-value chemicals is evaluated. The advantages and challenges of biocatalysis in drug discovery and development are also reviewed.

Well-Defined Phosphine-Free Manganese(II)-Complex-Catalyzed Synthesis of Quinolines, Pyrroles, and Pyridines

Summary: In this study, a simple, phosphine-free, and inexpensive catalytic system based on a manganese(II) complex was reported for synthesizing important N-heterocycles from amino alcohols and ketones. Control experiments, kinetic studies, and DFT calculations were conducted to support a plausible reaction mechanism. Potential intermediates in the catalytic cycle were detected using ESI-MS analysis, leading to the proposal of a metal-ligand cooperative mechanism.

Convenient and Reusable Manganese-Based Nanocatalyst for Amination of Alcohols

Summary: The development of new sustainable nanocatalytic systems for green chemical synthesis is a growing area in chemical science. A reusable heterogeneous N-doped graphene-based manganese nanocatalyst for selective N-alkylation of amines with alcohols is described. Mechanistic studies illustrate that the catalytic reaction follows a domino dehydrogenation-condensation-hydrogenation sequence of alcohols and amines with the formation of water as the sole by-product. The scope of the reaction is extended to the synthesis of pharmaceutically important N-alkylated amine intermediates, and the catalyst showed long-term performance and retained its activity after several recycling experiments.

Zinc Stabilized Azo-anion Radical in Dehydrogenative Synthesis of N-Heterocycles. An Exclusively Ligand Centered Redox Controlled Approach

Summary: The study presented a ligand-centered redox-controlled approach using a Zn(II)-stabilized azo-anion radical complex as a catalyst for efficient dehydrogenation of various N-heterocycles. The catalyst was found to be compatible with the cascade synthesis of N-heterocycles through dehydrogenative coupling of alcohols with other suitable coupling partners. Mechanistic investigation revealed that the dehydrogenation reactions proceed via a one-electron hydrogen atom transfer pathway and involve exclusive involvement of ligand-centered redox events with zinc acting only as the template.

Ruthenium-catalysed chemoselective alkytation of nitroarenes with alkanols

Summary: The alkylation of nitroarenes with alkanols catalyzed by a phosphinesulfonate chelated ruthenium complex [Ru-1] exhibits diverse reactivity and chemoselectivity depending on the acidic-basic conditions. This catalyst can promote the formation of secondary and tertiary anilines under neutral or acidic conditions, while enabling N-monoalkylation of nitroarenes with primary alkanols under alkaline conditions through a concerted approach. Mechanistic studies showed that the O-H activation of alkanols assisted by the sulfonate ligand is crucial in the novel reactivity observed under alkaline-free conditions.
