Metabolic engineering of microorganisms for L-alanine production

Summary: L-alanine is widely used in chemical, food, and medicine industries, with industrial production mainly relying on the enzymatic process using petroleum-based L-aspartic acid. Alternative routes for L-alanine production from renewable biomass via microbial fermentation have been explored. Despite the natural ability of many microorganisms to produce L-alanine, using native strains is limited by low yields and productivities.

Fusion of Formate Dehydrogenase and Alanine Dehydrogenase as an Amino Donor Regenerating System Coupled to Transaminases

Summary: In this study, a self-sufficient bifunctional enzyme was developed by genetically fusing alanine dehydrogenase (AlaDH) and formate dehydrogenase (FDH) for the recycling of amino donor and cofactor. The enzyme was coupled to transaminases for efficient chemical synthesis. The system successfully synthesized vanillyl amine and demonstrated the ability of the multienzyme system to be reused.
