The Emerging Role of Ferroptosis in Various Chronic Liver Diseases: Opportunity or Challenge

Summary: Ferroptosis is a form of intracellular cell death that is iron-dependent and mediated by the accumulation of lipid peroxides. It has distinct characteristics such as the loss of lipid peroxidase repair activity, the presence of redox-active iron, and the oxidation of phospholipids-containing polyunsaturated fatty acids. Recent studies have shown its involvement in various liver diseases, but there is controversy regarding its mechanism and regulation in different liver cell types. This review summarizes the current understanding of ferroptosis in chronic liver disease and highlights its potential as a therapeutic option.

Toxicity of Bisphenol A and its Replacements in the Mice Leydig Cells In Vitro

Summary: The aim of this study was to investigate the potential impacts of bisphenol A (BPA) and its analogues BPB, BPF, and BPS on mice TM3 Leydig cells, focusing on basal cell viability parameters such as metabolic activity, cell membrane integrity, and lysosomal activity after 48-h exposure. The study also evaluated the effects of bisphenols on ROS production, gap junctional intercellular communication (GJIC), and testosterone secretion. The results showed that activity started at 10 pg/ml of bisphenols after 48-h exposure, and significant ROS production was observed at 5 and 10 pg/ml. Moreover, a significant inhibition of GJIC was confirmed at 10, 25, and 50 pg/ml.

In vitro and in silico assessment of GPER-dependent neurocytotoxicity of emerging bisphenols

Summary: Through molecular docking and molecular dynamics simulation, the effects of eight bisphenol compounds (BPs) on G protein-coupled estrogen receptor (GPER) were evaluated and confirmed using IMR-32 cells. The study found that the target BPs significantly increased reactive oxygen species (ROS) production, decreased cell viability, activated apoptosis-related proteins expression, and increased apoptosis rate of IMR-32 cells. The MD simulation also demonstrated the binding conformations and dynamic changes of GPER-BPs, suggesting the activation of GPER by these BPs.

Early-life exposure to bisphenol A induces dysregulation of lipid homeostasis by the upregulation of SCD1 in male mice

Summary: Exposure to Bisphenol A (BPA) is associated with an increased prevalence of obesity-related metabolic syndrome. This study investigated the mechanisms of BPA-induced adipogenesis in preadipocytes and the effects of BPA exposure on lipid and glucose homeostasis using cell and animal models. The results showed that BPA exposure increased lipid content in adipocytes and caused a NAFLD-like phenotype in mice. RNA-Seq analysis revealed dysregulation in multiple biological processes, such as lipid metabolism and inflammation. Importantly, abnormal expression of SCD1 and APOD was found in association with BPA exposure and obesity.

Determination of multiple bisphenol analogues and their metabolites in human serum by liquid chromatography tandem mass spectrometry

In this study, a method involving dispersive solid-phase extraction and LC/MS was used to investigate the contamination levels of 28 precursor bisphenols and 9 major metabolites in serum. The results showed that BPA was the dominant contaminant, and BPA bis-(beta-D-glucuronide) may be a more important metabolite of BPA. Negligible potential health risks to blood donors were observed.

The results showed that BPA exposure increased levels of TG and TC in serum and liver, down-regulated the expression of liver fatty acid oxidation-related genes, and up-regulated the expression of fatty acid synthesis-related genes. Increased protein levels of mTOR and p-CRTC2 suggested the involvement of the mTOR/CRTC2/SREBP-1 pathway. Overall, these findings suggest that gestational BPA exposure can impact lipid metabolic disorders in later life.
