Critical endpoints of PFOA and PFOS exposure for regulatory risk assessment in drinking water: Parameter choices impacting estimates of safe exposure levels

Summary: USEPA has issued drinking water interim health advisories (iHA) for PFOA and PFOS, lowering the iHA values from 70 ppt to 0.04-0.2 ppt. This review addresses the key steps in the iHA derivation and suggests changes for higher confidence in the iHA derivation, such as reevaluation of critical endpoint data and modeling changes for PFAS elimination. Weaknesses in cancer epidemiological data are also discussed for reevaluation in the drinking water regulation process.

Evaluation of Per- and Polyfluoroalkyl Substances (PFAS) In Vitro Toxicity Testing for Developmental Neurotoxicity

Summary: Per- and polyfluoroalkyl substances (PFAS) are widely found commercial chemicals, but only a limited number of them have associated hazard identification data. This study used new approach methods (NAMs) to generate in vitro screening data for 160 PFAS in order to provide information on developmental neurotoxicity (DNT). The results showed that a subset of PFAS perturb neurodevelopmental processes in vitro and suggest focusing future studies of DNT on PFAS with certain structural feature descriptors.

Summary: The EPAs of Denmark, Sweden, Norway, Germany, and the Netherlands have proposed a ban on the use of toxic industrial chemicals PFAS due to their harmful effects on human health and biodiversity. Denmark, the first EU country to ban PFAS, plans to establish groundwater parks to protect drinking water. They also call on the EU to list more toxic substances on the Stockholm Convention.

Summary: The existence of thousands of PFAS with evidence of adverse health effects has raised the need for better understanding of PFAS toxicity beyond one-chemical-at-a-time approaches. The zebrafish model has enabled rapid assessment and comparison of PFAS compounds, leading to significant advances in PFAS research. This review focuses on assessing the toxicity and potential modes of action of PFAS using the zebrafish model, and recent data on diverse PFAS structures are enabling prioritization of compounds of concern.

Perfluorohexanesulfonic acid (PFHxS) induces oxidative stress and causes developmental toxicities in zebrafish embryos

Summary: In this study, zebrafish embryos were exposed to PFHxS and it was found that PFHxS accumulated in the embryos, leading to phenotypic abnormalities and disruption of cellular processes. Further analysis revealed that PFHxS affected glucose metabolism, lipid metabolism, and oxidative stress, resulting in increased reactive oxygen species, lipid accumulation, and cell cycle arrest. These findings highlight the potential toxicities of PFHxS.

Prenatal exposure to legacy PFAS and neurodevelopment in preschool-aged Canadian children: The MIREC cohort

Dietary Perfluorohexanoic Acid (PFHxA) Exposures in Juvenile Zebrafish Produce Subtle Behavioral Effects across Generations

Summary: Ubiquitous anthropogenic contaminants of concern, per- and polyfluoroalkyl substances (PFAS) are frequently detected in the environment and human populations around the world. Diet is a predominate route of human exposure, and PFAS are frequently measured in food. However, the current amount of data to support safety assessments of these alternatives is not yet sufficient. The present study investigated the effects of a 42-day dietary exposure to 1, 10, or 100 ng/g PFHxA in juvenile zebrafish. In conclusion, PFHxA exposure induced abnormal adult anxiety behaviors that persisted transgenerationally, and abnormal behavior was also observed in the F1 generation. This study highlights the need for further investigation of this and other alternative short-chain PFAS.

Physiological and transcriptomic effects of hexafluoropropylene oxide dimer acid in Caenorhabditis elegans during development

Drinking Water-Associated PFAS and Fluoroethers and Lipid Outcomes in the GenX Exposure Study

The results showed that PFOS and PFNA were associated with higher total and non-HDL cholesterol levels, especially in older adults. A rapid assessment bioaccumulation screening (RABS) study design for emerging per-and polyfluoroalkyl substances in mice exposed to industrially impacted surface water

Summary: The shift away from PFOS and PFOA production in the past two decades has led to the release of complex mixtures of emerging PFAS with unknown bioaccumulation potential and toxicology. The lack of available chemical standards often limits the prioritization of research on these emerging PFAS in complex mixtures. This study developed a rapid assessment method to prioritize emerging PFAS in an environmentally derived mixture based on their potential for mammalian bioaccumulation. The findings show that certain PFAS, such as Nafion BP2, Hydro-EVE, PFO(4)DA, and PFO(5)DoA, have a higher bioaccumulation potential, supporting previous studies on human serum from communities with contaminated drinking water.
