Profile and actual transmissibility of Carbapenem resistance genes: Intracellular and extracellular DNA in hospital wastewater

Hospital wastewaters (HWWs) are significant reservoirs for antibiotic resistance genes, but the resistomes in HWWs, particularly CRGs, are not well characterized. This study comprehensively characterized the profile and transmissibility of extracellular and intracellular CRGs in HWWs for the first time.

Profiling of intracellular and extracellular antibiotic resistance genes in municipal wastewater treatment plant and their effluent-receiving river

Summary: The presence of antibiotic resistance genes (ARGs) and heavy metal resistance genes (MRGs) in the municipal wastewater treatment plant and its adjacent river sediments has been investigated. Results showed that internal DNA had higher absolute abundances of ARGs and MRGs compared to external DNA in the wastewater treatment plant, while the opposite was observed in river sediments. Aminoglycoside resistance genes were found to be the most abundant ARGs, while the zntA gene was one of the most abundant MRGs. The presence of the mobile genetic element intI1 was positively correlated with the proliferation of resistance genes in both internal and external DNA. This widespread distribution of ARGs, MRGs, and MGEs provides insights into the transmission routes and highlights the importance of monitoring and mitigating their dissemination.

Synergistic removal of pharmaceuticals and antibiotic resistance from ultrafiltered WWTP effluent: Free-floating ARGs exceptionally susceptible to degradation

Summary: In order to protect the environment and human health, it is necessary to remove antibiotic resistance genes (ARGs) and persistent pharmaceuticals from WWTP effluent before reuse. This study proposes a combination of UV/H2O2 treatment, GAC and zeolite sorption to efficiently remove free-floating extracellular DNA (exDNA) and a wide range of pharmaceuticals. The combined approach significantly reduces the levels of ARGs and removes most pharmaceutical residues, making the effluent suitable for reuse.

Identification of critical control points for antibiotic resistance discharge in sewers

Summary: Disrupting the spread of clinically relevant antibiotic resistance genes (ARGs) is crucial for the success of the One Health strategy. This study focuses on monitoring wastewater feeds of selected urban drainage areas, and identifies hospitals and retirement homes as major sources of high levels of ARGs in wastewater. The monitoring approach includes qPCR-based quantifications, flow volume-based analyses, and antibiotic resistance analyses. The study suggests that local actions at critical control points can prevent contamination of larger volumes of wastewaters in a cost-effective manner, compared to central actions at wastewater treatment plants (WWTPs). The implementation of a polluter-pays principle is recommended for this monitoring strategy.
