PPARa/ACOX1 as a novel target for hepatic lipid metabolism disorders induced by per- and polyfluoroalkyl substances: An integrated approach

Summary: This study aimed to explore the link between PFAS exposure and NAFLD and investigate the molecular mechanisms involved. Cross-sectional analyses using NHANES data showed a significant association between PFAS exposure and hepatic steatosis/NAFLD. Further in vivo and in vitro experiments confirmed that ACOX1-mediated oxidative stress contributed to mitochondrial compromise and lipid accumulation in PFOA/PFOS-exposed mouse hepatocytes. Additionally, the study found that hepatic ACOX1 exhibited a response to short-term exposures of long-chain PFAS in human hepatocyte spheroids.

Human biomonitoring (HBM) based on PFAS determination in serum samples is a suitable strategy for assessing population exposure and risk. The paper presents the evaluation process and results of quality assurance and quality control (QA/QC) measures for PFAS analysis in serum, showing the successful establishment of a network of laboratories with high analytical comparability in 12 European countries.

Summary: Perfluoroalkyl ether carboxylic acids (PFECAs), including substances like PFO4DA and PFO5DoDA, have been found in polluted regions in both surface water and volunteer blood samples. The study showed that PFO5DoDA had a longer serum half-life and stronger hepatic bioaccumulation compared to PFO4DA, contributing to its stronger hepatotoxicity. Chronic exposure to these compounds led to weight gain, disrupted stress signals, and disturbed glucose and lipid metabolism in the liver.
