Mre11 exonuclease activity promotes irreversible mitotic progression under replication stress

Yoshitami Hashimoto et al.

Summary: The study reveals a novel role of Mre11 exonuclease activity in promoting the initial progression of mitotic entry under replication stress and suggests that it facilitates the processing of stalled replication forks, leading to irreversible mitotic progression and replisome disassembly.

DDK: The Outsourced Kinase of Chromosome Maintenance

Peter J. Gillespie et al.

Summary: To ensure genetic stability, cells must duplicate their complement of chromosomes prior to cell division. This process involves replicating chromosomal DNA and re-establishing the chromatin environment. The duplication of chromatid pairs must remain coupled until cell division to ensure equal segregation of genetic material. Cyclin-dependent kinases (CDK) control the regulation of chromosome duplication. CDK also activates Dbf4-dependent kinase (DDK), which facilitates DNA replication activation and coordination with chromatin re-establishment and chromatid coupling. This review discusses the outsourcing of these activities by CDK to DDK to ensure chromosome maintenance during cell division.

Spotlight on the Replisome: Aetiology of DNA Replication-Associated Genetic Diseases

Roberto Bellelli et al.

Summary: Cellular proliferation and differentiation are dependent on regulated DNA replication by the replisome, a large machine in eukaryotes. Mutations in replisome components can lead to genetic disorders characterized by developmental abnormalities, reduced organismal growth, and heightened tumour predisposition. Understanding the molecular genetics and pathogenesis of these disorders is crucial for developing targeted therapies.

The role of DDK and Treslin-MTBP in coordinating replication licensing and pre-initiation complex formation

Ilaria Volpi et al.

Summary: Treslin/Ticrr is essential for DNA replication initiation and forms a complex with MTBP. DDK and CDK activities cooperate to regulate the interaction of Treslin-MTBP, which is crucial for selecting replication origins. This study provides insights into the mechanisms behind replication initiation regulation.

Mechanisms of Vertebrate DNA Interstrand Cross-Link Repair

Daniel R. Semlow et al.

Summary: This review explores the role of ICLs in DNA repair, the relationship between diseases like Fanconi anemia and cancer with ICL repair, the chemical diversity of ICLs, and how cells unhook these interstrand cross-links. The review also highlights new unhooking strategies, advancements in the structural analysis of the Fanconi anemia pathway, and insights into how cells choose between different ICL repair pathways. Gaps in our understanding of these DNA repair pathways are emphasized throughout the discussion.

Replication initiation: Implications in genome integrity

Yo-Chuen Lin et al.

Summary: During every cell cycle, billions of nucleotides need to be accurately duplicated within hours. Cells regulate the replication event by assembling essential factors during the G1 phase to establish the pre-replicative complex, and during S phase the replication process is tightly coupled with the DNA repair system. Defects and errors in replication must be recognized and halted by DNA damage response and checkpoint signaling pathways, highlighting the critical coordination of these processes to achieve genomic integrity.

Replication of the Mammalian Genome by Replisomes Specific for Euchromatin and Heterochromatin

Jing Zhang et al.

Summary: During DNA replication, replisomes have different replication schedules in different chromatin environments, and their replication process can be influenced by replication stress. Currently, there is limited understanding of the determinants of replisome stalling vs. continued progression, but it has been discovered that replisomes with different activities exist in different stages and chromatin environments.

Ongoing replication forks delay the nuclear envelope breakdown upon mitotic entry

Yoshitami Hashimoto et al.

Summary: The study investigated the restart of stalled replication forks during mitosis using Xenopus egg extracts. Stalled forks could restart before nuclear envelope breakdown with excess dCTP, but efficiency decreased after NEB. The timing of NEB was delayed by ongoing forks rather than stalled forks, dependent on Wee1/Myt1 kinase activities.
