Liquid demixing in elastic networks: Cavitation, permeation, or size selection?

Pierre Ronceray et al.

Summary: In this study, the researchers investigate the mechanical interactions between phase-separated liquid condensates and surrounding elastic networks in cells. Through theoretical analysis, they propose the existence of three thermodynamically stable droplet phases, including macroscopic droplets that either cavitate or permeate the network, and mesh-size-limited microdroplets. The study also suggests the possibility of yet-unobserved droplet phases in the cytoplasm and nucleoplasm.

Sequence-dependent surface condensation of a pioneer transcription factor on DNA

Jose A. Morin et al.

Summary: This study investigates the nature of transcription factor condensates and demonstrates that transcription factors can undergo phase separation at high concentrations and form condensates on DNA at low concentrations. These condensates are shown to form on DNA as a type of surface condensation and are correlated with DNA sequence, thus explaining the size and position control of surface condensates.

RNA at the surface of phase-separated condensates impacts their size and number

Audrey Cochard et al.

Summary: This study presents a method to form RNA-containing condensates in living cells. Programmable tools are used to recruit a specific RNA species and control the formation and dissolution of condensates. The recruitment of RNA changes the material properties of condensates, and the size of condensates scales with RNA surface density.

Membrane surfaces regulate assembly of ribonucleoprotein condensates

Wilton T. Snead et al.

Summary: This study reveals that membrane tethering plays a role in the assembly of ribonucleoprotein condensates and restricts their size by reducing protein and RNA diffusion. The interaction between ribonucleoprotein condensates and the endoplasmic reticulum affects the size of the condensates. The concentration and diffusion of membrane-bound complexes contribute to the formation and coarsening of condensates.

ALS-linked KIF5A ΔExon27 mutant causes neuronal toxicity through gain-of-function

Devesh C. Pant et al.

Summary: This study confirms the association between mutations in the human KIF5A gene and ALS. The specific mutation identified leads to the production of Delta Exon27, which is neurotoxic and causes neurodegeneration and disease progression at the cellular and organism levels, ultimately leading to the development of ALS.

Axonal mRNA translation in neurological disorders

Julie Qiaojin Lin et al.

Summary: Local protein synthesis (LPS) is increasingly recognized for its contribution to axon biology, with disruptions in LPS associated with various neurological disorders. Axons are exceptionally reliant on LPS due to their length and autonomy, which plays crucial roles in maintaining axonal structure and function, as well as establishing neuronal connectivity.

Cavitation controls droplet sizes in elastic media

Estefania Vidal-Henriquez et al.

Summary: The study presents a theoretical description of droplets growing in an elastic matrix, revealing that large droplets only form when cavitation occurs, stabilizing the size and position of smaller droplets and increasing in density with faster cooling. The model also indicates how adjusting cooling protocols and nucleation site density affects droplet size distribution.

Designer membraneless organelles sequester native factors for control of cell behavior

Mikael Garabedian et al.

Summary: The subcellular compartmentalization of macromolecules is essential for controlling biochemical reactions by increasing flux and preventing inhibitory interactions. We have developed a synthetic membraneless organelle platform that can control endogenous cellular activities by sequestering and insulating native proteins. By relocalizing targeted enzymes to synthetic condensates, we are able to efficiently regulate cellular behaviors such as proliferation, division, and cytoskeletal organization, and demonstrate strategies for releasing cargo from condensates to switch cells between functional states. These synthetic organelles offer a powerful and versatile approach to controlling cell decision-making in various model systems, with broad applications in cellular engineering.

Mammalian Neuronal mRNA Transport Complexes: The Few Knowns and the Many Unknowns

Elsa C. Rodrigues et al.

Summary: The article discusses how mRNA is sorted into different packages and coupled to motor proteins for transport. It remains unclear whether all mRNAs are transported by the same machinery, or if different adaptors or motors are used for different transcripts. While there is indirect evidence for the involvement of proteins in mRNA localization, relatively little is known about the essential activities required for the actual transport process.
