Real-Time Objective Evaluation of the Ischemic Stroke through pH-Responsive Fluorescence Imaging

Summary: Rapid and comprehensive assessment of ischemic stroke (IS) is crucial for clinicians to provide appropriate treatment. Currently, IS assessment relies on computed tomography and magnetic resonance imaging, along with clinical symptoms and contraindication inquiries. However, these methods cannot diagnose pathology or assess the microenvironment in real-time. This study develops a pH-responsive fluorescent liposomal probe for visualizing the degree of IS based on acid microenvironments. The fluorescence imaging with this probe provides a new method for objectively evaluating IS severity and offers insights into the relationship between acidic microenvironments and IS progression.

Near-Infrared Ratiometric Hemicyanine-Based pH Fluorescence Probe with Bone Targetability for Monitoring Bone Resorption

Summary: A near-infrared ratiometric hemicyanine-based pH fluorescence probe was designed and synthesized for reliable and safe monitoring of bone resorption in orthodontic treatment. In vitro and in vivo experiments demonstrated that the probe had high sensitivity, excellent photostability, reversibility, and strong resistance to interference. The probe could detect bone resorption through fluorescence imaging and quantitative monitoring of pH. Therefore, it has great clinical potential for real-time monitoring of bone resorption in orthodontic treatment and as a reference for other bone resorption-related diseases.

A pH-sensitive carbonic anhydrase IX-targeted near-infrared probe for fluorescent sensing and imaging of hypoxic osteosarcoma

Summary: Osteosarcoma (OS) is a common and deadly carcinoma of the skeletal system. Detecting the tumor early and implementing effective interventional therapies can improve limb salvage and overall survival rates. Carbonic anhydrase IX (CA IX) is an important functional protein in tumor acidification and is considered a potential biomarker. This study developed a pH-sensitive near-infrared imaging probe called ABSi-783 by combining the CA IX inhibitor ABSi with IR783. The probe has high selectivity for osteosarcoma imaging, can detect pH changes in vitro, and has demonstrated potential for targeted diagnosis and surgical treatment monitoring of osteosarcoma.

Lysosome-targeted high resolution near-infrared fluorescent probe for low physiological pH imaging in vivo

It possesses high-resolution characteristics, good selectivity, and excellent photostability. The probe can conveniently and sensitively track the lysosomal pH fluctuation in living cells. Visualizing the Interplay of Lipid Droplets and Protein Aggregates During Aging via a Dual-Functional Fluorescent Probe

Summary: This study describes a dual-functional fluorescent probe, LW-1, for simultaneously imaging lipid droplets (LDs) and protein aggregates (PAs) and reveals their interplay during aging. The results demonstrate that the level of PAs in intestine tissues increases with aging, accompanied by the buildup of LDs. These findings highlight the importance of LDs and PAs during the aging process in intestine tissues.

Ratiometric fluorescent carbon dots for enantioselective sensing of L-lysine and pH discrimination in vivo and in vitro

Summary: In this study, a ratiometric fluorescent probe with wavelength-resolved dual target sensing capacity was successfully synthesized, allowing simultaneous recognition of different target objects. The novel fluorescent carbon dots exhibited discrete dual-emission bands and served as fluorescent nanoprobes for the chiral recognition of Lysine and the ratiometric detection of pH. Confocal imaging experiments demonstrated real-time monitoring of L-Lysine and pH fluctuations in biological systems using this nanoprobe.

Quinolizinium-based tunable pH fluorescent probes for imaging in live cells

Summary: A new type of quinolizinium-based fluorescent probes has been developed for cell imaging with tunable pH responsiveness. These probes exhibit turn On fluorescence response upon acidification through modulation of photoinduced electron transfer (PET) from the N,N-dialkyl aniline substituent to the quinolizinium core skeleton. They demonstrate tunable emission properties and pH responsiveness with pKa ranging from <2.1 to 6.1, thanks to the steric effect of N,N-dialkyl anilines. Successful application for in vivo pH sensing in lysosome in live cells has been achieved.
