An endoplasmic reticulum-specific ratiometric fluorescent probe for imaging esterase in living cells

Summary: Esterase is primarily distributed in the endoplasmic reticulum (ER) and often overexpressed in cancer cells. To solve the limitations of existing probes, an ER-specific ratiometric fluorescent probe (ER-EST) is designed for detecting esterase in living cells. The ER-EST shows a ratiometric and red-shifted emission from 435 to 560 nm after hydrolysis by esterase, and it has a low detection limit and good biocompatibility.

Peroxynitrite activatable double spiral ring derived fluorescent probe for imaging of inflammation and drug-induced liver injury

Summary: Researchers have developed two unique fluorescent probes that can monitor the real-time changes of peroxynitrite (ONOO-) in the process of inflammation and drug-induced liver injury (DILI), providing a powerful tool for studying disease progression and prognosis. These probes have been successfully applied to detect endogenous and exogenous ONOO- in living cells and zebrafish, and confirmed the production of ONOO- mediated by CYP450 in the process of APAP-induced DILI. Additionally, in vivo imaging using the probe revealed the upregulation of ONOO- in inflammatory sites and damaged liver, which was confirmed by HE staining. These results highlight the great potential of the probe in the early diagnosis of DILI and its contribution to drug screening.

A Reversible Fluorescent Probe for In Situ Monitoring Redox Imbalance during Mitophagy

A Reversible Fluorescent Probe for In Situ Monitoring Redox Imbalance during Mitophagy

Summary: Mitophagy is a process of degrading damaged and dysfunctional mitochondria that is closely associated with H2O2-related redox imbalance and pathological processes. A reversible fluorescent probe (M-HP) was developed for real-time imaging of H2O2-related redox imbalance. The probe showed rapid response and high sensitivity to the H2O2/GSH redox cycle, with a detection limit of 17 nM for H2O2. In living cells, M-HP demonstrated excellent reversibility and mitochondrial targeting for imaging H2O2 fluctuation. The results revealed an increase in mitochondrial H2O2 under lipopolysaccharide stimulation and a decrease in H2O2 following combined treatment with rapamycin, suggesting significant suppression of oxidative stress after enhancement of mitophagy.
