On the Utility of Chemical Strategies to Improve Peptide Gut Stability

Summary: The inherent susceptibility of peptides to enzymatic degradation in the gastrointestinal tract is a major challenge in oral peptide drug development. This study systematically analyzed the gut stability of different peptides and discussed medicinal chemistry strategies to improve peptide gut stability. It was found that cyclotides were the only scaffold class that could resist gastrointestinal degradation, and site-specific alterations were effective in improving stability. The importance of standardized test conditions and defined protocols for cross-study comparison was also highlighted.
