Identification of deleterious variants of uncertain significance in BRCA2 BRC4 repeat through molecular dynamics simulations

Summary: In this study, we proposed a method based on molecular dynamics simulation to classify variants of uncertain significance in cancer predisposition genes. By examining the impact of these variants on protein structure stability, we were able to differentiate them into deleterious and tolerated variants. This approach showed high sensitivity and specificity in classifying missense variants and can be applied to other cancer predisposition genes.

BRCA2 BRC missense variants disrupt RAD51-dependent DNA repair

Summary: This study identified three BRCA2 VUS located in the BRC repeat region and investigated their impact on genome stability. The S1221P and T1980I mutations were found to compromise the cellular response to chemotherapeutics and ionizing radiation, and showed deficits in fork protection. The T1346I mutation, on the other hand, was fully functional. The correct classification of BRCA2 VUS is important as pathogenic variants can impact cancer risk and guide therapy selection during cancer treatment.

Improved RAD51 binders through motif shuffling based on the modularity of BRC repeats

Summary: The study focused on the role of RAD51-interacting modules within BRCA2, creating chimeric repeats to measure binding to RAD51. Surprisingly, certain shuffled module combinations showed stronger binding than natural repeats, with weak correlation to affinities of natural repeats. The strongest chimera, BRC8-2, showed improved binding to RAD51 and functioned in preventing the formation of nuclear RAD51 foci in human cells after ionizing radiation.
