A feedback loop between heterochromatin and the nucleopore complex controls germ-cell-to-oocyte transition during Drosophila oogenesis

H/ACA snRNP-dependent ribosome biogenesis regulates translation of polyglutamine proteins

Summary: The H/ACA snRNP complex plays a crucial role in oocyte specification by promoting ribosome biogenesis and pseudouridylation of rRNA. By reducing ribosome levels, translation of specific mRNAs encoding polyglutamine-containing proteins, including differentiation factors, is decreased during terminal differentiation. Ribosomes are also enriched at CAG repeats within transcripts during oogenesis. Increasing TOR activity can rescue GSC differentiation defects in H/ACA snRNP complex-depleted germlines, while TOR inhibition leads to reduced levels of polyglutamine-containing proteins. These findings highlight the role of ribosome biogenesis and levels in controlling stem cell differentiation.

Summary: In sexually reproducing animals, a subset of RNAs expressed during early oogenesis undergo regulated degradation, failure to remove these RNAs results in oocyte dysfunction and death. Key regulators of oogenesis through targeted degradation of specific RNAs include the RNA-degrading Super Killer complex and No-Go Decay factor Pelota. Targeted RNAs are enriched for cytidine sequences bound by the polypyrimidine tract binding protein Half pint, orchestrating a germ cell-to-maternal transition giving rise to the maternal contribution to the zygote.
