PINK1/TAX1BP1-directed mitophagy attenuates vascular endothelial injury induced by copper oxide nanoparticles

Summary: This study found that CuONPs exposure disrupted mitochondrial dynamics in vascular endothelial cells through oxidative stress-dependent manner, as evidenced by increased mitochondrial fission and accumulation of fragmented mitochondria. Inhibition of mitochondrial fission aggravated CuONPs-induced mtROS production and cell death. Furthermore, mitochondrial fission led to the activation of PINK1-mediated mitophagy, and inhibition of mitophagy repressed CuONPs-induced toxicity. TAX1BP1 was identified as the primary receptor linking ubiquitinated mitochondria with autophagosomes, and its knockdown aggravated CuONPs-induced cell death. Urolithin A, a mitophagy activator, promoted mtROS clearance and removal of damaged mitochondria induced by CuONPs exposure both in vitro and in vivo.

Cardioinotropic Effects in Subchronic Intoxication of Rats with Lead and/or Cadmium Oxide Nanoparticles

Some of the toxic and genotoxic effects could be partially or fully normalized when intoxication occurred alongside the administration of a bioprotective complex.
