MLKL-Driven Inflammasome Activation and Caspase-8 Mediate Inflammatory Cell Death in Influenza A Virus Infection

Xuqiu Lei et al.

Summary: Influenza A virus (IAV) induces multiple types of cell death, including necroptosis, apoptosis, and pyroptosis. MLKL promotes inflammasome activation and IL-1 beta processing in IAV-infected macrophages, while caspase-8 coordinates the maturation and secretion of IL-1 beta in the absence of the MLKL-inflammasome axis. MLKL and caspase-8 serve as redundant mechanisms for inflammatory cell death in response to IAV infection.

ZBP1-dependent inflammatory cell death, PANoptosis, and cytokine storm disrupt IFN therapeutic efficacy during coronavirus infection

Rajendra Karki et al.

Summary: This study found that ZBP1 induced during coronavirus infection limits the efficacy of IFN therapy by driving inflammatory cell death and lethality. Inhibiting ZBP1 activity may improve the efficacy of IFN therapy for COVID-19 and other infections and inflammatory conditions where IFN-mediated cell death and pathology occur.

ADAR1 mutation causes ZBP1-dependent immunopathology

Nicholas W. Hubbard et al.

Summary: The RNA-editing enzyme ADAR1 inhibits innate immune activation and pathology by disrupting the duplex structure of endogenous double-stranded RNA species. Alteration of the Z-DNA-binding domain (ZBD) of ADAR1 leads to activation of ZBP1, causing autoinflammatory pathology.

ADAR1 averts fatal type I interferon induction by ZBP1

Huipeng Jiao et al.

Summary: Mutations of the ADAR1 gene cause chronic activation of type I interferon responses, leading to severe diseases. The interaction between ADAR1 and another protein called ZBP1 prevents IFN activation. Deficiency or mutation of ZBP1's Z alpha domains reduces expression of IFN-stimulated genes and prevents early postnatal lethality in mice with mutated ADAR1 Z alpha domain.

ADAR1 prevents autoinflammation by suppressing spontaneous ZBP1 activation

Richard de Reuver et al.

Summary: The RNA-editing enzyme ADAR1 plays an important role in limiting the accumulation of immunostimulatory dsRNA. Reduced ADAR1 activity can lead to severe inflammatory diseases, while complete loss of ADAR1 activity is embryonically lethal and causes autoinflammation. ADAR1 not only prevents overactivation of MDA5 and PKR, but also inhibits the spontaneous activation of ZBP1.

Molecular and structural aspects of gasdermin family pores and insights into gasdermin-elicited programmed cell death

Ayesha Zahid et al.

Summary: Pyroptosis is a highly inflammatory and lytic type of programmed cell death initiated by inflammasomes, with recent studies linking a family of pore-forming proteins called gasdermins (GSDMs) to this process. GSDMs have characteristic features of forming pores in the plasma membrane by releasing N-terminal fragments that can bind to acidic lipids, leading to disruption of cellular homeostasis and cell death induction. Recent studies have also revealed the structural and molecular aspects of autoinhibition, as well as the functional consequences of gasdermin-induced membrane permeabilization.

Synergism of TNF-α and IFN-γ Triggers Inflammatory Cell Death, Tissue Damage, and Mortality in SARS-CoV-2 Infection and Cytokine Shock Syndromes

Rajendra Karki et al.

Summary: The combination of TNF-alpha and IFN-gamma induces inflammatory cell death characterized by PANoptosis, which can be inhibited to protect mice from the pathological effects and death caused by COVID-19. Blocking the cytokine-mediated inflammatory cell death signaling pathway may benefit patients with COVID-19 or other infectious and autoinflammatory diseases by limiting tissue damage/inflammation.

ADAR1 restricts ZBP1-mediated immune response and PANoptosis to promote tumorigenesis

Rajendra Karki et al.

Summary: The study uncovers the role of ADAR1 in suppressing ZBP1-mediated PANoptosis and promoting tumorigenesis. Additionally, the use of IFN-gamma and NEI KPT-330 regresses melanoma in a ZBP1-dependent manner, providing important insights for cancer therapeutic strategies.

Viral dosing of influenza A infection reveals involvement of RIPK3 and FADD, but not MLKL

Teodora Oltean et al.

Summary: The study found that RIPK3 plays an important role in protection against influenza A virus, but this protection is only apparent within a limited dose range of IAV challenge. The protective outcome is independent of RIPK3 kinase activity and MLKL, suggesting that the platform function of RIPK3 is crucial for protection. FADD-dependent apoptosis also has a crucial additional effect in protection against IAV infection.

AIM2 forms a complex with pyrin and ZBP1 to drive PANoptosis and host defence

SangJoon Lee et al.

Summary: Inflammasomes are essential for innate immune defence, sensing pathogens and inducing cell death. They can detect multiple molecular patterns released by live pathogens during infection, activating multiple inflammasome sensors simultaneously. The study found that AIM2 regulates innate immune sensors pyrin and ZBP1 to drive inflammatory signalling and a form of inflammatory cell death known as PANoptosis, providing host protection during infections with specific pathogens.

Extracellular Nucleic Acids Present in the Candida albicans Biofilm Trigger the Release of Neutrophil Extracellular Traps

Magdalena Smolarz et al.

Summary: Neutrophils utilize various antimicrobial mechanisms, including the production of neutrophil extracellular traps (NETs) through NETosis, to combat invading pathogens. Fungal infections, particularly those caused by Candida albicans, can be defended against using NETosis strategy. The ability of C. albicans to form biofilms and release extracellular nucleic acids plays a significant role in developing drug resistance and promoting NET release by inducing reactive oxygen species (ROS) production and cell migration.
