Molecular Aspects and Therapeutic Implications of Herbal Compounds Targeting Different Types of Cancer

Aditi Sharma et al.

Summary: Due to genetic changes, cancer remains a major cause of death. Anticancer research faces challenges in targeting healthy cells while addressing the importance of target function. Cancer cells undergo various mutations and abnormalities that affect the efficacy of anticancer drugs. With low clinical success rates, finding alternative herbal-based therapies to attenuate resistance and protect against chemotherapy side effects is crucial. This review updates the understanding of common cancer types and presents key compounds that may enhance cancer treatment efficacy.

MD simulation-based screening approach identified tolvaptan as a potential inhibitor of Eg5

Jomon Sebastian et al.

Summary: We discovered tolvaptan as a new Eg5 inhibitor using molecular dynamics simulation-based virtual screening. Tolvaptan exhibited stable binding to Eg5 and inhibited the growth of HeLa cells through mitotic block. It also bound to goat brain tubulin and overlapped with colchicine in terms of binding location. Tolvaptan could be a prospective anticancer agent, either alone or in combination with other antimitotic drugs.

Review Metastasis

Stefanie Gerstberger et al.

Summary: Most cancer-associated deaths are caused by metastasis, which is still being studied for a better understanding and more effective treatment. Metastasis involves the acquisition of traits for dissemination, variable dormancy, and colonization of distant organs. Clonal selection, dynamic cell state transitions, and immune environment co-option are driving factors for successful metastasis. This review highlights the main principles of metastasis and presents emerging opportunities for developing better therapies.

Consequences of Glucose Enriched Diet on Oncologic Patients

David Gonzalez-Flores et al.

Summary: Malignant tumors have increased glucose utilization and uptake, so clinical trials are evaluating inhibitors of glycolytic metabolism. Antiglycolytic drugs can enhance chemotherapy and radio-induced cell death. Glucose influences the levels and activation of pro-apoptotic and anti-apoptotic proteins, leading to regulation of the apoptotic machinery. Dietary modifications can impact tumor growth by altering nutrient availability in the tumor microenvironment. Recent studies show that increasing key minerals in the diet can affect the efficacy of cancer therapies, underscoring the importance of considering diet and nutrition during cancer research and treatment.

Cancers Attributable to Modifiable Risk Factors: A Road Map for Prevention

Giulia Collatuzzo et al.

Summary: The implementation of cancer preventive strategies relies on evidence from cancer epidemiology, identifying risk factors and estimating their prevalence. However, even when cancer is caused by modifiable factors, prevention through their control is not always feasible. We reviewed associations between modifiable risk factors and cancer, highlighting both successful and unsuccessful examples of cancer control and emphasizing the incomplete understanding of avoidable causes of cancer.

Direct and indirect effects of tubulin post-translational modifications on microtubule stability: Insights and regulations

Julia Baer et al.

Summary: Microtubules possess changeable and tightly controlled properties, which are mediated by a variety of tubulin posttranslational modifications (PTMs) that either directly alter the MT lattice structurally or indirectly change MT interaction partners. Understanding the reversal and regulation of PTMs on MTs is crucial in deciphering their roles in cellular functions. Advances in microscopy techniques and live-sensors have greatly facilitated the study of PTM interactions and effects.
