Fast and sensitive GCaMP calcium indicators for imaging neural populations

Yan Zhang et al.

Summary: In this study, fast and sensitive GCaMP-type indicators were developed and optimized through large-scale screening and structure-guided mutagenesis. The resulting jGCaMP8' sensors, with ultra-fast kinetics and high sensitivity, can track large populations of neurons.

Hippocampal-hypothalamic circuit controls context-dependent innate defensive responses

Jee Yoon Bang et al.

Summary: This study reveals a new mechanism for experience-dependent, top-down control of innate defensive behaviors in animals, showing that the anterior hypothalamic nucleus is the key brain region involved, with the hippocampus transmitting information through this pathway to control memory and escape behaviors in prey.

Regulation of body weight and food intake by AGRP neurons during opioid dependence and abstinence in mice

Brenton T. Laing et al.

Summary: There is an interaction between opioids and energy deficit, with opioids attenuating feeding behavior and activation of ARC(AGRP) neurons during opioid dependence reducing weight loss without causing weight gain.

Coordination of escape and spatial navigation circuits orchestrates versatile flight from threats

Weisheng Wang et al.

Summary: This study demonstrates that cholecystokinin-expressing cells in the hypothalamic dorsal premammillary nucleus play a crucial role in controlling context-specific escape behaviors in mice, unlike other nuclei implicated in flight. The dorsal premammillary nucleus is able to influence spatial navigation and escape circuits, leading to versatile flight behaviors.

Translational PET applications for brain circuit mapping with transgenic neuromodulation tools

Matthew A. Boehm et al.

Summary: Transgenic neuromodulation tools, such as chemogenetic and optogenetic systems, have revolutionized neuroscience by allowing targeted manipulation of neuronal activity with unprecedented specificity. These tools enable mapping of neuroanatomical projection sites and linking cellular manipulations with brain circuit functions and behavior, expanding knowledge of the nervous system in preclinical models. New strategies for implementing and monitoring transgenic tools, such as positron emission tomography (PET) imaging techniques, are needed to track the location and function of chemogenetic receptors and opsins in vivo for safe and effective use in translational research and potential clinical applications.

Hypothalamic control of interoceptive hunger

Justin N. Siemian et al.

Summary: The study reveals the distinct roles of different hypothalamic neuronal populations in regulating internal hunger states and food intake behavior. The manipulation of specific neuronal circuits can evoke varied effects on feeding behaviors, shedding light on potential therapeutic targets for eating disorders.

Lateral hypothalamic LEPR neurons drive appetitive but not consummatory behaviors

Justin N. Siemian et al.

Summary: Research shows that a subpopulation of LH GABAergic neurons expressing leptin receptors specifically drives appetitive behaviors in mice, while their ablation does not affect weight gain and food intake. Both groups of neurons in the LH can modulate reward-related behaviors, but only LHVGAT neurons impact feeding, and LHLEPR neurons can discriminate conditioned cues in Pavlovian paradigms.
