A Drug-Drug Cocrystal of Dihydromyricetin and Pentoxifylline

Summary: Drug-drug cocrystals, which can regulate physicochemical properties of individual drugs and produce synergistic therapeutic effects, have gained attention in the pharmaceutical industry. In this study, a novel drug-drug cocrystal was prepared using a slurry method, resulting in synchronous and sustained release of two drugs with different aqueous solubility. The cocrystal exhibited reduced hydrophilicity and demonstrated a synergistic anti-cancer effect in vitro.

Formation Thermodynamics of Carbamazepine with Benzamide, Para-Hydroxybenzamide and Isonicotinamide Cocrystals: Experimental and Theoretical Study

Summary: Thermodynamic parameters for the formation of cocrystals of carbamazepine with structurally related coformers were determined using experimental and computational techniques. The relationship between solubility products and solubility values of the coformers was found, and the relationship between the entropy term and molecular volume of cocrystal formation was revealed. A new method for assessing cocrystal stability based on hydrogen bond propensity analysis was proposed. The experimental and theoretical results on the thermodynamic parameters were in good agreement.

The correlation equations connecting the melting points of co-crystals and individual components have been obtained. An approach for evaluating the ideal solubility of co-crystals based on the ideal solubility of one of the co-crystal individual components has been developed.
