Bacteria-Driven Tumor Microenvironment-Sensitive Nanoparticles Targeting Hypoxic Regions Enhances the Chemotherapy Outcome of Lung Cancer

Summary: A new Bifidobacterium infantis (Bif)-based biohybrid (Bif@PDA-PTX-NPs) was designed to deliver polydopamine (PDA)-coated paclitaxel nanoparticles (PTX-NPs) to tumor tissues. Bif@PDA-PTX-NPs maintained the toxicity of PTX and the hypoxic homing tendency of Bif, significantly inhibiting the growth of A549 xenografts in nude mice and prolonging the survival of tumor-bearing mice.

Summary: Oral squamous cell carcinoma (OSCC) in non-smoking and non-drinking individuals is different from traditional head and neck squamous cell carcinoma (HNSCC). The incidence of this type of cancer is increasing, especially in young females, and the tongue is the most affected subsite. Although gene mutations and unique epigenomic profiles have been identified, there is no definitive genetic cause. Other possible causes include chronic dental trauma, microbiome abnormalities, marijuana consumption, and genetic disorders.

Bacteria, with their potential for non-pathogenicity through genetic manipulation and virulence factors, are being explored as viable options for cancer therapy, despite challenges such as cytotoxic effects and incomplete cancer cell lysis.

Summary: Local hypoxia is a common feature of solid tumors. In this study, a biocompatible bacteria/nanoparticles biohybrid platform was developed to target breast tumors. The results showed that the biohybrid was able to effectively colonize hypoxic tumors, increasing drug accumulation and reducing toxic side-effects.
