Halogenated class of oximes as a new class of monoamine oxidase-B inhibitors for the treatment of Parkinson's disease: Synthesis, biochemistry, and molecular dynamics study

Development of Isopropyl-Tailed Chalcones as a New Class of Selective MAO-B Inhibitors for the Treatment of Parkinson?s Disorder

Summary: Thirteen isopropyl chalcones have been synthesized and evaluated for their inhibitory activity against monoamine oxidase (MAO). The compounds exhibited stronger inhibition against MAO-B than MAO-A. Compound CA4 showed the most potent inhibition against MAO-B with an IC50 value of 0.032 μM, similar to CA3 (IC50 = 0.035 μM), and had high selectivity index values for MAO-B over MAO-A. Compound CA10 showed the strongest inhibition against MAO-A with an IC50 value of 0.310 μM and effectively inhibited MAO-B (IC50 = 0.074 μM).

Aldoxime- and hydroxy-functionalized chalcones as highly potent and selective monoamine oxidase-B inhibitors

Summary: A panel of chalcone derivatives were evaluated for their inhibitory activities, and HC6 was found to be the most potent inhibitor of MAO-B. HC3 and ACE7 also showed high inhibitory activity. These findings suggest that HC3 and HC6 hold promise as potential candidates for neurological disorder treatments.

Conjugated Dienones from Differently Substituted Cinnamaldehyde as Highly Potent Monoamine Oxidase-B Inhibitors: Synthesis, Biochemistry, and Computational Chemistry

Summary: Fifteen multiconjugated dienones were synthesized and evaluated for their inhibitory activities against MAO-A and MAO-B. The compounds showed potent and highly selective inhibition of MAO-B. MK6 and MK12 exhibited the most effective inhibition of MAO-B, with high selectivity indices. The study concluded that these compounds have potential as neurodegenerative disorder treatments.

CHB3 and CHF3 showed the most potent inhibitory activity against MAO-B and were identified as reversible competitive inhibitors. CHB3 was considered a potential candidate for treating neurological disorders such as Parkinson's disease.
