A comprehensive biophysical and theoretical study on the binding of dexlansoprazole with human serum albumin

Anna Tanuja Safala Bodapati et al.

Summary: The present study investigates the interaction between the new generation proton pump inhibitor dexlansoprazole (DLP) and human serum albumin (HSA). Fluorescence and UV-visible spectroscopy were used to quantify the interaction and determine parameters such as quenching constant, binding constant, and thermodynamic parameters. The results show that DLP and HSA interact with a hypochromic effect observed in absorption studies. The binding constants obtained from fluorescence studies indicate a collisional-type quenching mechanism. The study also confirms the binding of DLP in subdomain IIA of HSA through site marker competitive displacement studies and molecular docking.

Structural aspects of formetanate hydrochloride binding with human serum albumin using spectroscopic and molecular modeling techniques

Shweta Singh et al.

Summary: This study investigated the binding characteristics of Formetanate Hydrochloride (FMT) with Human serum albumin (HSA) using experimental and computational methods. The results showed that the binding of FMT to HSA is spontaneous and entropy driven. FMT binds to specific domains and subdomains of HSA and is stabilized by hydrophobic interactions. Additionally, molecular dynamics simulations revealed minor adjustments in FMT's conformation within the binding site, and computational results were consistent with experimental findings.

Chuanying Zhang et al.

Summary: Brazilin, hematoxylin, and hematein are polyphenols with similar structures and rich biological activities, and they have protective effects against oxidative degradation of human serum albumin. The biological effects of these polyphenols are mainly influenced by their binding to protein, and fluorescence experiments indicate that they bind to protein through a static quenching mechanism, causing some conformational changes. These findings provide insights into the role of molecular structure in the protective effects and binding abilities of polyphenols against protein oxidative damage.

Structural and Dynamical Differences in the Spike Protein RBD in the SARS-CoV-2 Variants B.1.1.7 and B.1.351

Nisha Bhattarai et al.

Summary: The ongoing novel coronavirus pandemic has led to new variants like B.1.1.7 and B.1.351, which show increased transmissibility and infectivity. Molecular dynamics simulations revealed rearranged hydrogen bond networks and new bonds between the virus spike protein RBD and host receptor ACE2 due to mutations. The B.1.351 variant exhibited higher flexibility in the RBD loop segment, but the RBD-ACE2 complex showed greater stability, which could contribute to increased infectivity of the mutated variants.
