Engineered periosteum-diaphysis substitutes with biomimetic structure and composition promote the repair of large segmental bone defects

Lili Yu et al.

Summary: This study investigates the repair of large segmental bone defects using tissue-engineered periosteum-diaphysis substitutes. By co-culturing bone marrow mesenchymal stem cells (BMSCs) and endothelial progenitor cells (EPCs) on electrospun silk fibroin (SF) fibrous membranes, an artificial periosteum cambium was created to mimic the cellular composition and microstructure of the native periosteum. The biomimetic periosteum-diaphysis substitutes promoted the repair of critical-size bone defects in rabbits and prevented the growth of fibrous tissues, reducing the occurrence of nonunion.

Natural micropatterned fish scales combing direct osteogenesis and osteoimmunomodulatory functions for enhancing bone regeneration

Di Qin et al.

Summary: Surface topography of fish scales promotes bone regeneration by directing cell behavior and modulating the osteoimmune microenvironment. The anisotropic ridged micropatterns and mineralized collagen on the scales induce macrophage polarization and stimulate anti-inflammatory cytokine secretion. In addition, they enhance osteogenic differentiation of mesenchymal stem cells and increase bone formation. The findings suggest that fish scales can be a potential implant material for bone repair.

Bone ECM-inspired biomineralization chitin whisker liquid crystal hydrogels for bone regeneration

Lin Li et al.

Summary: This study presents a novel bone extracellular matrix (ECM)-inspired biomineralization chitin whisker liquid crystal (LC) hydrogel, which exhibits chiral nematic LC state and viscoelasticity similar to natural bone ECM. Through guided biomineralization, the LC hydrogel accurately mimics the mineralization process of natural bone, resulting in the formation of organic-inorganic composite materials with a bone ECM-like microenvironment. These biomineralization chitin whisker LC hydrogels show enhanced biomineralization, protein adsorption, and osteogenesis abilities, providing a promising approach for bone regeneration.

Lu Wang et al.

Summary: In this study, silk fibroin/nano-hydroxyapatite/graphene oxide composite scaffolds with directional channels were prepared and systematically studied. The directed channel scaffolds showed better connectivity, porosity, degradation rate for osteogenesis, and ability to induce bone-like apatite. They were also more beneficial to the adhesion and proliferation of bone marrow mesenchymal stem cells, and exhibited stronger biological activity and osseointegration ability. Therefore, this material has great potential in repairing bone tissue defects.

The bone microenvironment invigorates metastatic seeds for further dissemination

Weijie Zhang et al.

Summary: Metastasis is often considered the final step of tumor progression, but recent studies show that many metastases are initiated by the further spread of existing metastases. Through experiments, it was found that the bone microenvironment can facilitate cancer cells to further metastasize and establish secondary metastases in multiple organs. This metastasis-promoting effect is driven by epigenetic reprogramming that confers stem cell-like properties on disseminated cancer cells.

Enhanced compressive strengths and induced cell growth of 1-3-type BaTiO3/PMMA bio-piezoelectric composites

Yufei Tang et al.

Summary: Barium titanate (BaTiO3) is used as a bone implant material due to its piezoelectric properties and ability to promote cell growth with hydroxyapatite. A method combining directional freeze casting and self-solidification of bone cement was explored to improve the piezoelectric properties of 1-3-type BaTiO3/PMMA biopiezoelectric composites with a lamellar structure. Lamellar BaTiO3 layers and dendritic ceramic bridges on BaTiO3 pore walls enhance the compressive strength and elastic modulus of the composite, while polarized lamellar BaTiO3 can induce osteoblasts to grow along the layers.
