Summary: Frontotemporal dementia (FTD) is a common form of dementia that affects the frontal and temporal lobes of the brain. The etiology of FTD is complex, with both familial and sporadic cases. Deficiency in progranulin (PGRN) has been linked to microglial-associated neuroinflammation, TDP-43 pathology, and lysosomal dysfunction.
