Cell-Free DNA Promotes Thrombin Autolysis and Generation of Thrombin-Derived C-Terminal Fragments

Summary: Cell-free DNA (cfDNA) is a major component of neutrophil extracellular traps (NETs), which play a critical role in innate immune response to infection. Recent studies have shown that NETs have procoagulant activity and can trigger thrombin generation through the intrinsic pathway of coagulation. In vitro experiments have shown that DNA binding can lead to thrombin autolysis and generation of thrombin-derived C-terminal peptides (TCPs), which can bind to NETs and provide protection against degradation.

Summary: The canonical NLRP3 inflammasome participates in NETosis under sterile conditions by regulating NLRP3 and ASC protein levels, while genetic ablation of NLRP3 signaling impairs NET formation. Inhibition of NLRP3 decreases NETosis, and NLRP3 deficiency results in reduced NET density in thrombi in a mouse model of deep vein thrombosis.

Summary: This study aimed to compare the efficacy of TAC and CsA regimens as initial treatments for patients with PM/DM-ILD. Results showed that PSL plus TAC treatment may achieve a better short-term PFS rate, while PSL plus CsA treatment had a slight advantage in the OS rate at 52 weeks. Additionally, both groups showed significant improvements in pulmonary function tests, with no treatment discontinuations due to adverse events.
