Single-cell RNA-seq methods to interrogate virus-host interactions

Summary: Tumor associated macrophages (TAMs) are abundant subpopulations in several solid cancers and their number is linked to poor clinical outcomes. Stromal cells, such as cancer associated fibroblasts (CAFs), play a role in TAM recruitment, survival, and reprogramming. Single cell-RNA sequencing (sc-RNA seq) technologies have provided new insights into TAMs and CAFs, revealing their phenotypical and functional programs. This mini-review focuses on recent discoveries in sc-RNA seq regarding TAM and CAF identity and their crosstalk in the tumor microenvironment (TME) of solid cancers.

Cellular and molecular insights into incomplete immune recovery in HIV/AIDS patients

Summary: Highly active antiretroviral therapy (ART) can effectively inhibit HIV replication and restore immune function. However, a significant number of patients do not achieve satisfactory increases in CD4(+) T cell counts, leading to incomplete immune reconstitution or immunological nonresponse (INR). INR patients have an increased risk of clinical progression and higher mortality rates. The mechanisms underlying INR are still unclear, despite extensive research. This review aims to explore the cellular and molecular factors contributing to incomplete immune reconstitution.

Monocyte-derived macrophages contain persistent latent HIV reservoirs

Summary: The development of persistent cellular reservoirs of latent HIV in myeloid cells is a critical obstacle to viral eradication. This study develops a quantitative viral outgrowth assay and highly sensitive T cell detection assays to assess the frequency of latent HIV in monocytes of virologically suppressed individuals with HIV. The results show that myeloid cells meet the definition of a clinically relevant HIV reservoir and emphasize the importance of including myeloid reservoirs in efforts towards an HIV cure.

Identification of Potential Diagnostic Genes of HIV-Infected Immunological Non-Responders on Bioinformatics Analysis

Upregulation of IFN-stimulated genes persists beyond the transitory broad immunologic changes of acute HIV-1 infection

Summary: Chronic immune activation during HIV-1 infection is characterized by persistent elevation of a narrow set of interferon-stimulated genes and innate cytokines. Genes such as MX1, IFI27, ISG15 are upregulated and correlated with plasma HIV-1 RNA, TNF-a, and CXCL10 cytokine levels during acute and chronic infection. In contrast, genes involved in cellular immune responses, such as CD8A, are upregulated during acute infection before returning to near pre-infection levels in chronic infection. These findings suggest the potential for targeted interventions to restore healthy immune homeostasis in people living with HIV-1.

Methods: This review focuses on the use of single-cell RNA sequencing and other single-cell analysis technologies to uncover the heterogeneity of pancreatic ductal adenocarcinoma in different cellular subpopulations. Results and Conclusion: The application of single-cell RNA sequencing has revealed associations between disease progression and heterogeneity in the pancreatic tumor microenvironment, which could have implications for future treatment strategies.
