Inhibitors of UHRF1 base flipping activity showing cytotoxicity against cancer cells

Stefano Ciaco et al.

Summary: UHRF1, a nuclear multi-domain protein overexpressed in various human cancer types, can be inhibited by two new compounds, AMSA2 and MPB7, which prevent UHRF1/DNMT1 interaction during DNA replication, reduce overall DNA methylation, and induce cell death specifically in cancer cell lines with high levels of UHRF1. These compounds show promise as anti-cancer drugs targeting UHRF1 due to their specificity and effectiveness.

Unprecedented Combination of Polyketide Natural Product Fragments Identifies the New Hedgehog Signaling Pathway Inhibitor Grismonone

Michael Grigalunas et al.

Summary: Pseudo-natural products (pseudo-NPs) are novel bioactive compounds that are formed by combining fragments of natural product (NP) in new ways. Previously, pseudo-NPs were created by combining fragments from unrelated NPs, but this study shows that combining fragments from related NPs can also lead to novel chemotypes with unique bioactivity.

Recent Advances in Natural Product-Based Hybrids as Anti-Cancer Agents

Eleni Sflakidou et al.

Summary: Cancer is a leading cause of death worldwide. Natural products have been used for cancer treatment since ancient times, but their limitations in pharmacokinetics, selectivity, and efficacy against drug-resistant cancer cells hinder their clinical application. Conjugation of natural products with other bioactive molecules can overcome these limitations and develop novel bifunctional drugs for cancer therapy.

Chemical diversity, medicinal potentialities, biosynthesis, and pharmacokinetics of anthraquinones and their congeners derived from marine fungi: a comprehensive update

Mohamed Sebak et al.

Summary: Marine fungi are important sources of structurally unique secondary metabolites that have promising potential as substitutes for current therapeutics. Anthraquinones, a group of polyketides derived from filamentous fungi, have diverse biological activities, including anticancer, antimicrobial, antioxidant, and anti-inflammatory effects. This review comprehensively summarizes the research conducted over the past 20 years on 208 structurally diverse anthraquinones and their derivatives isolated from different species of marine-derived fungi, along with their reported bioactivity. The study also presents recent insights into their biosynthetic routes and evaluates their drug-likeness and pharmacokinetics properties.

Drug resistance and Cancer stem cells

Yuan Li et al.

Summary: Therapy resistance is a significant challenge in treating cancer patients, with intrinsic and acquired factors contributing to the development of resistance. CSCs play a key role in therapy resistance, as they are inherently resistant to anticancer treatments and can easily adapt to changes in the tumor microenvironment.
