Targeting lactate metabolism and glycolytic pathways in the tumor microenvironment by natural products: A promising strategy in combating cancer

Leila Kooshki et al.

Summary: Anticancer drugs are limited in their effectiveness due to toxicity, side effects, high costs, and resistance. Cancer's ability to adapt to different signaling pathways and evade immune responses makes it a complex health issue. Recent research highlights the critical role of lactate in cancer development, emphasizing the importance of targeting lactate metabolism/transporters for prevention and management of tumors. Natural secondary metabolites emerge as promising alternative compounds with high efficacy and fewer side effects, targeting lactate-related pathways to combat cancer.

Novel dual-targeting c-Myc inhibitor D347-2761 represses myeloma growth via blocking c-Myc/Max heterodimerization and disturbing its stability

Ruosi Yao et al.

Summary: A novel dual-targeting c-Myc inhibitor D347-2761 has been identified, which can effectively inhibit the growth of multiple myeloma, induce apoptosis, and overcome drug resistance when used in combination with bortezomib. Additionally, this inhibitor inhibits myeloma growth and infiltration by promoting c-Myc protein degradation.

RIP140 inhibits glycolysis-dependent proliferation of breast cancer cells by regulating GLUT3 expression through transcriptional crosstalk between hypoxia induced factor and p53

Valentin Jacquier et al.

Summary: Glycolysis plays a crucial role in cancer cell proliferation, and RIP140 is a transcriptional co-regulator involved in mammary tumorigenesis. Our study shows that RIP140 deficiency promotes glycolysis-dependent breast tumor growth and decreases the transcription of the glucose transporter gene GLUT3 by inhibiting the activity of the transcription factor HIF-2 alpha in cooperation with p53. Interestingly, RIP140 expression is associated with good prognosis in breast cancer patients with low GLUT3 expression, low HIF-2 alpha expression, and high p53 expression.

Functional inhibition of c-Myc using novel inhibitors identified through hot spot targeting

Ashutosh Singh et al.

Summary: Protein-protein interactions play important roles in both healthy and diseased states. This study identified hot spot residues within the disordered c-Myc/Myc-associated factor X interface using computational methods, and demonstrated through biophysical and cell-based experiments that small molecules can interact with these hot spots to deregulate c-Myc activity and exhibit potent anti-tumor effects against c-Myc-expressing cancer/stem cells.

Gene Expression Profiling and Protein Analysis Reveal Suppression of the C-Myc Oncogene and Inhibition JAK/STAT and PI3K/AKT/mTOR Signaling by Thymoquinone in Acute Myeloid Leukemia Cells

Belal Almajali et al.

Summary: This study found that thymoquinone (TQ) from Nigella sativa inhibits the activation of JAK/STAT and PI3K/AKT signaling pathways, leading to downregulation of c-Myc expression in HL60 leukemia cells. This potential inhibition of proliferation and induction of apoptosis highlights the therapeutic potential of TQ in treating leukemia.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Trimetazidine alone or in combination with gemcitabine and/or abraxane decreased cell viability, migration and ATP levels and induced apoptosis of human pancreatic cells

Vedat Atli Sekeroglu et al.

Summary: The study demonstrates that trimetazidine in combination with abraxane and/or gemcitabine induces significant cytotoxicity and apoptosis in human pancreatic cancer cells, while also reducing cell migration and ATP levels. These findings suggest that a combination strategy based on inhibition of fatty acid oxidation and anticancer drugs may be more effective in the treatment of pancreatic cancers.

Effect of Heat Stress on Bovine Mammary Cellular Metabolites and Gene Transcription Related to Amino Acid Metabolism, Amino Acid Transportation and Mammalian Target of Rapamycin (mTOR) Signaling

Lin Fu et al.

Summary: This study utilized metabolomics technology to investigate the changes in intracellular metabolites and gene expression related to amino acid metabolism, transportation, and the mTOR signaling pathway in bovine mammary epithelial cells under heat stress (HS) conditions. The findings suggest that HS significantly alters intracellular amino acid metabolism and maintains milk protein synthesis by promoting amino acid transportation and activating the mTOR signaling pathway.

ATIC inhibits autophagy in hepatocellular cancer through the AKT/FOXO3 pathway and serves as a prognostic signature for modeling patient survival

Hao Zhang et al.

Summary: The study identified the correlation between autophagy-related genes and the prognosis of hepatocellular carcinoma patients, and established a reliable prognostic model based on 12 genes. This model can effectively predict patients' survival time, reflecting their immune characteristics and status. Furthermore, ATIC inhibits autophagy and promotes the progression of liver cancer through the AKT/FOXO3 signaling pathway.
