High-dimensional isotomics, part 1: A mathematical framework for isotomics

Summary: Molecules exist in different forms, called isotopologues, with varying numbers of isotopic substitutions. The concentration of these isotopologues in a sample, known as the isotome, encodes information about the sample's history. Recent advances enable the measurement of constraints on the sample's isotome, but there is a challenge in relating these constraints to the abundances of all possible isotopologues. In this study, a mathematical framework is developed to understand the relationship between different measurements of a sample's isotome.

Accuracy and precision of ESI-Orbitrap-IRMS observations of hours to tens of hours via reservoir injection

Summary: Orbitrap-IRMS has been successfully applied to high-precision isotope ratio measurements of various compounds, but certain ratios require long experimental times to achieve desired precision. This study characterizes the performance of Orbitrap-IRMS for long duration measurements and explores the challenges of inter-measurement stability. Results show high precision for most ratios but limited accuracy due to inter-measurement drift. The study demonstrates the viability of Orbitrap-IRMS for extended measurements of challenging properties.

Methods and limitations of stable isotope measurements via direct elution of chromatographic peaks using gas chromotography-Orbitrap mass spectrometry

Summary: This study examines the performance of the Thermo Scientific Orbitrap mass spectrometer for isotope ratio measurements in a gas chromatograph with direct elution. The results suggest that this method is suitable for compounds with large position-specific, molecular, or multiply-substituted isotopic anomalies and for compounds that produce strong molecular ions. Precision can be improved by targeting ions with high mass spectral intensities, optimizing the number and range of masses admitted, and minimizing the resolution settings while separating relevant isobars.

Quantifying Isotopologue Reaction Networks (QIRN): A modelling tool for predicting stable isotope fractionations in complex networks

Summary: Natural-abundance stable isotope compositions are important tools for understanding complex processes. This article presents a new isotope modelling software tool called QIRN, which combines computational strategies used in metabolic modeling with natural isotope fractionations. QIRN treats isotopic properties as distributions of discrete isotopologues, enabling models of reaction networks with unprecedented complexity. QIRN can model any physical, chemical or biological process and has a diverse range of applications. It reproduces outputs from previous models and predicts isotopic anomalies measured in nature.

Exploring frontiers of orbitrap performance for long transients

Summary: Experiments on a modified Orbitrap Exploris instrument have demonstrated that a resolving power in excess of 2,000,000 can be achieved even in a compact instrument when appropriate tolerance and tuning requirements are met. Improved pumping design in a new Orbitrap allows isotopic resolution of intact monoclonal antibodies, with longer transients improving signal-to-noise ratios towards single-charge resolution levels in charge detection mass spectrometry. Further steps for enhancing instrumentation are discussed.
