Summary: This study found a significant positive correlation between CXCR7 expression and CAF activation markers in colonic tissues and transgenic mice. RNA sequencing revealed an increase in miR-146a-5p and miR-155-5p levels in CXCR7-overexpressing CRC cells and their exosomes. Furthermore, these CRC cell-derived miRNAs could be uptaken by CAFs via exosomes and promote their activation, leading to enhanced invasive capacity of CRC cells and pro-metastatic switch. The activation of CAFs by miR-146a-5p and miR-155-5p also facilitated tumor formation and lung metastasis of CRC in vivo. Therefore, these findings provide new insights into CXCR7-mediated CRC metastasis and suggest potential biomarkers and therapeutic targets for inhibiting CRC metastasis.

Gastric cancer-derived exosomal miR-519a-3p promotes liver metastasis by inducing intrahepatic M2-like macrophage-mediated angiogenesis

Summary: This study found that miR-519a-3p expression levels in exosomes were higher in gastric cancer liver metastasis (GC-LM) patients, and high expression indicated a worse prognosis. GC-derived exosomes mainly accumulated in the liver and were internalized by intrahepatic macrophages. Further research revealed that miR-519a-3p activated the MAPK/ERK pathway by targeting DUSP2, inducing M2-like polarization of macrophages and promoting angiogenesis and intrahepatic premetastatic niche formation. These findings suggest that miR-519a-3p plays a critical role in the interaction between primary GC cells and intrahepatic macrophages.
