Overall survival with first-line atezolizumab in combination with vemurafenib and cobimetinib in BRAFV600 mutation- positive advanced melanoma (IMspire150): second interim analysis of a multicentre, randomised, phase 3 study

Paolo A. Ascierto et al.

Summary: The phase 3 IMspire150 study showed improved investigator-assessed progression-free survival with atezolizumab, vemurafenib, and cobimetinib in patients with BRAFV600 mutation-positive melanoma. However, the interim overall survival analysis did not demonstrate a significant improvement in overall survival compared to the control group.

DHX38 restricts chemoresistance by regulating the alternative pre-mRNA splicing of RELL2 in pancreatic ductal adenocarcinoma

Zeru Li et al.

Summary: The study identified a new intron retention gene in pancreatic ductal adenocarcinoma, which is regulated by the upstream splicer DHX38. The reduction of DHX38 expression leads to the retention of the fourth intron of RELL2, causing nonsense-mediated mRNA decay and inhibiting apoptosis of pancreatic cancer cells, ultimately promoting drug resistance. This research provides a potential therapeutic target for pancreatic ductal adenocarcinoma.

RAF-MEK-ERK pathway in cancer evolution and treatment

Rahim Ullah et al.

Summary: The RAF-MEK-ERK signaling cascade is a well-characterized MAPK pathway that plays a critical role in cell proliferation and survival. Genetic alterations in this pathway are highly prevalent in human cancers and can lead to uncontrolled cell growth and tumor formation. The crosstalk between the RAF-MEK-ERK axis and other signaling pathways further enhances its proliferative potential in human cancers.

Retinitis pigmentosa-linked mutation in DHX38 modulates its splicing activity

Mina Obuca et al.

Summary: This study analyzed the role of RNA helicase DHX38 in retinitis pigmentosa (RP). The findings showed that DHX38 is associated with key splicing factors, regulates alternative splicing and promotes the usage of cryptic splice sites. The RP-linked substitution G332D modulates DHX38's splicing activity.

FOXD1-dependent RalA-ANXA2-Src complex promotes CTC formation in breast cancer

Yufei Long et al.

Summary: In this study, a FOXD1-dependent RalA-ANXA2-Src complex that promotes CTC formation via activating ERK1/2 signal in BC was discovered. FOXD1 may serve as a prognostic factor in evaluating BC metastasis risks. This signaling cascade is druggable and effective for overcoming early stages of BC CTC formation.

Anticancer drug resistance: An update and perspective

Ruth Nussinov et al.

Summary: Driver mutations play a critical role in cancer initiation and progression, with drug resistance being a common challenge in pharmacological treatment. The emergence of drug resistance can be attributed to a variety of mechanisms, such as mutations occurring in the same or different proteins, and in the same or parallel pathways. The complexity of genetic alterations and tumor microenvironment changes poses challenges in predicting drug resistance and requires further research in precision medicine.

Summary: BRAF and KRAS mutations play a significant role in NSCLC by activating the RAS/RAF/MEK/ERK signaling pathway. Current approved therapies include BRAF and MEK inhibitors, while combination with other drugs may enhance therapeutic effects. Further research is needed to explore optimal drug combinations for treating lung cancer patients.

HIF-1α switches the functionality of TGF-β signaling via changing the partners of smads to drive glucose metabolic reprogramming in non-small cell lung cancer

Yiwei Huang et al.

Summary: The study reveals that TGF-beta plays a dual role in regulating glycolysis and cell proliferation in non-small cell lung cancer. HIF-1 alpha is identified as a critical factor in this process, potentially serving as a biomarker or therapeutic target for TGF-beta mediated cancer progression. The research findings provide insight into the metabolic reprogramming mechanisms in tumor cells.

Mex3a promotes oncogenesis through the RAP1/MAPK signaling pathway in colorectal cancer and is inhibited by hsa-miR-6887-3p

Haixia Li et al.

Summary: The study demonstrated that Mex3a plays a significant role in colorectal cancer (CRC) by regulating the RAP1GAP/MEK/ERK/HIF-1α pathway. Mex3a was identified as a prognostic factor for overall survival in CRC patients, and knockdown of Mex3a inhibited tumor cell migration, invasion, and proliferation. Furthermore, hsa-miR-6887-3p was found to downregulate Mex3a expression and inhibit CRC tumorigenesis both in vitro and in vivo.
