Spatial multiomics map of trophoblast development in early pregnancy

Anna Arutyunyan et al.

Summary: In this study, a spatially resolved multiomics single-cell atlas of the human maternal-fetal interface was generated and used to identify possible transcription factors involved in trophoblast invasion.(4,3,5) The transcriptomes of the final cell states of trophoblast invasion were defined, and cell-cell communication events contributing to trophoblast invasion and placental bed giant cell formation were predicted. The dual role of interstitial EVTs and endovascular EVTs in mediating arterial transformation during early pregnancy was also modeled.

Trophectoderm differentiation to invasive syncytiotrophoblast is promoted by endometrial epithelial cells during human embryo implantation

Peter T. Ruane et al.

Summary: Embryo attachment to endometrial epithelium leads to the formation of multinuclear syncytiotrophoblast, which breaches the epithelial layer. This process is crucial for successful implantation and involves gene networks that are potential targets for improving implantation success rates.

Spatiotemporal transcriptomic atlas of mouse organogenesis using DNA nanoball-patterned arrays

Ao Chen et al.

Summary: Spatially resolved transcriptomic technologies enable us to study complex biological processes such as mammalian embryogenesis. However, current methods have limitations in resolution, gene capture, and field of view, which hinders their systematic application to large and three-dimensional mid- and late-gestation embryos. In this study, we developed Stereo-seq, a spatially enhanced resolution omics-sequencing method, by combining DNA nanoball-patterned arrays and in situ RNA capture. We used Stereo-seq to generate the mouse organogenesis spatiotemporal transcriptomic atlas, MOSTA, which provides single cell resolution and high sensitivity for mapping the kinetics and directionality of transcriptional variation during mouse organogenesis. By utilizing this atlas, we investigated the molecular basis of spatial cell heterogeneity and cell fate specification in developing tissues like the dorsal midbrain. Our panoramic atlas will facilitate in-depth research into long-standing questions about normal and abnormal mammalian development.

Maternal physiology and blastocyst morphology are correlated with an inherent difference in peri-implantation human embryo development

Deirdre M. Logsdon et al.

Summary: This retrospective study found that patient characteristics such as body mass index, age, follicle-stimulating hormone: luteinizing hormone ratio on menstrual cycle day 3, antral follicle count on menstrual cycle day 3, and antimullerian hormone level on menstrual cycle day 3, as well as blastocyst morphological grade, were correlated with the developmental potential of the peri-implantation embryo. Blastocysts from older patients developed fewer epiblast cells compared to blastocysts from younger patients. These findings are important in understanding factors that influence preimplantation embryo developmental potential.

Trophoblast uptake of DBP regulates intracellular actin and promotes matrix invasion

Ankana Ganguly et al.

Summary: Early pregnancy is characterized by elevated levels of vitamin D binding protein (DBP). The presence of DBP in the placenta and its association with malplacentation disorders like preeclampsia have been studied. Research shows that DBP plays a crucial role in trophoblast matrix invasion, intracellular actin-binding, and maintaining pregnancy health.

Naive stem cell blastocyst model captures human embryo lineage segregation

Ayaka Yanagida et al.

Summary: Researchers have developed a human embryo model called blastoid through self-organization, which can form blastocyst-like structures within a short period of time, displaying three tissue layers and mimicking natural blastocysts. Single-cell transcriptome analysis confirms the high fidelity of the blastoid model, providing a versatile and scalable experimental model for human embryo research.

Large-scale analysis of imprinting in naive human pluripotent stem cells reveals recurrent aberrations and a potential link to FGF signaling

Gal Keshet et al.

Summary: This study demonstrates that naive hPSCs exhibit high levels of non-random loss of imprinting, with a bias towards paternally methylated imprinting control regions. The extent of loss of imprinting is tightly correlated with FGF signaling, showing the importance of these events in disease modeling, development, and cell therapy.
