Loss of Functional SCO2 Attenuates Oxidative Stress in Diabetic Kidney Disease

Nehaben A. Gujarati et al.

Summary: The study found that increased oxidative stress in glomerular endothelial cells (GEnCs) contributes to early diabetic kidney disease, but the loss of functional SCO2 leading to reduced complex IV activity may have a protective effect on GEnCs.

Mitochondria-targeted drugs for diabetic kidney disease

Akira Mima

Summary: Diabetic kidney disease (DKD) is a common cause of chronic kidney disease (CKD) in the United States, with chronic hyperglycemia believed to be the primary cause. Recent research has highlighted the potential of targeting mitochondrial function as a novel therapeutic approach for DKD, such as improving mitochondrial dynamics to provide renal protection.

Klotho-derived peptide 6 ameliorates diabetic kidney disease by targeting Wnt/beta-catenin signaling

Xiaowen Chen et al.

Summary: The Klotho-derived peptide KP6 mimics the protective effects of Klotho in diabetic kidney disease, alleviating established proteinuria, glomerular hypertrophy, podocyte damage, glomerulosclerosis, and interstitial fibrotic lesions by blocking the Wnt/beta-catenin signaling pathway.

Lysophosphatidylcholine mediates fast decline in kidney function in diabetic kidney disease commentary

Kentaro Yoshioka et al.

Summary: In this study, a comprehensive metabolomic analysis was performed on patients with G3 stage DKD, and increased urinary LPC levels were identified in fast decliners. The increase in urinary LPC levels was associated with a decline in estimated glomerular filtration rate. A rat model of DKD also showed increased levels of LPC in urine and kidney tubulointerstitium as the disease progressed. In vitro studies confirmed that LPC induced lipotoxicity in proximal tubular cells. Therefore, LPC (16:0) and (18:0) may play a role in the fast progression of DKD and could be a target for novel therapeutic approaches.

Role of urinary H2O2, 8-iso-PGF2α, and serum oxLDL/β2GP1 complex in the diabetic kidney disease

Rani Sauriasari et al.

Summary: The study investigated the association between ROS and their modified products with eGFR in T2DM patients, showing that only H2O2 had a correlation with eGFR in the total study population. Multiple linear regression analysis revealed that significant factors for increased eGFR were H2O2, diastolic blood pressure, and female gender, while increased systolic blood pressure and age were significant factors affecting the decrease of eGFR. Furthermore, urinary H2O2 was found to correlate with serum oxLDL/beta(2)GPI complex in the total population, suggesting potential for further research on urinary H2O2 in early detection and novel therapies for diabetic kidney disease.

Summary: According to GBD 2019 data, diabetes mortality rates in individuals under 25 globally decreased from 1990 to 2019, with the lowest decline seen in low SDI countries. Providing better access to insulin and diabetes care is crucial for reducing early mortality rates.

Podocyte-specific Nlrp3 inflammasome activation promotes diabetic kidney disease

Khurrum Shahzad et al.

Summary: This study found that activation of the NLRP3 inflammasome in podocytes is both necessary and sufficient for diabetic kidney disease (DKD), indicating that podocytes have immune cell-like functions. Targeting NLRP3 may be a promising therapeutic approach for DKD.

Neutrophil Extracellular Traps Induce Glomerular Endothelial Cell Dysfunction and Pyroptosis in Diabetic Kidney Disease

Fengfan Zheng et al.

Summary: The study found increased deposition of NETs in the glomeruli of DKD patients and diabetic mice, leading to glomerulopathy and GECs injury. Degradation of NETs or knockout of specific genes can alleviate these conditions. In vitro experiments showed that NETs can induce pyroptosis in GECs, which can be inhibited by strengthening the GECs surface charge.

Summary: This study reveals that AKAP1 phosphorylates Larp1 via PKC signaling activation to decrease mtDNA replication, which accelerates mitochondrial dysfunction and podocyte injury in DKD.
