Engineering the Intestinal Lymphatic Transport of Oral Nanoparticles to Educate Macrophages for Cancer Combined Immunotherapy

In this study, MTG nanoparticles were engineered to encapsulate siSIRP alpha and pMUC1, which can educate macrophages via oral route and exert cooperative antitumor effects. Liposome-trimethyl chitosan nanoparticles codeliver insulin and siVEGF to treat corneal alkali burns by inhibiting ferroptosis

Summary: In this study, a new combination therapy using liposomes and trimethyl chitosan (TMC) for the codelivery of insulin (INS) and vascular endothelial growth factor small interfering RNA (siVEGF) to treat alkali-burned corneas was designed. The therapy effectively inhibited oxidative stress, inflammation, and VEGF expression in vitro and maintained corneal transparency, accelerated epithelialization, and inhibited corneal neovascularization (CNV) in vivo. Metabolomic analysis suggested that the therapeutic mechanism of siVEGF-TIL might be associated with the inhibition of the ferroptosis signaling pathway. Overall, siVEGF-TIL NPs could be a safe and effective therapy for corneal alkali burn.

Chitosan polyplexes: Energetics of formation and conformational changes in DNA upon binding and release

Summary: The energetics of chitosan (CS) polyplexes and the conformational stability of bound DNA were analyzed at pH 5.0 using ITC and HS-DSC, respectively. The CS-DNA binding mechanism was found to involve cooperative attachment of interacting CS ligands to the DNA matrix, as shown by the well-fitted McGhee-von Hippel model. The melting thermograms of polyplexes revealed the transformation of different conformational forms of bound DNA depending on the CS/DNA weight ratio.

GE11-modified carboxymethyl chitosan micelles to deliver DOX•PD-L1 siRNA complex for combination of ICD and immune escape inhibition against tumor

Summary: A dual-responsive carboxymethyl chitosan (CMCS) micelle functionalized with a targeting peptide GE11 (G-CMssOA) was developed for simultaneous delivery of PD-L1 siRNA and doxorubicin (DOX) in a complex form (D&P). The complex-loaded micelles (G-CMssOA/D&P) exhibited good physiologic stability and pH/reduction responsiveness, and enhanced intratumoral infiltration of CD4(+) and CD8(+) T cells, reduced Tregs (TGF-beta) and increased immune-stimulatory cytokine secretion (TNF-alpha). The combination of DOX-induced immunogenic cell death (ICD) and PD-L1 siRNA-mediated immune escape inhibition significantly improved antitumor immune response and suppressed tumor growth. This complex delivery strategy provides a novel approach for effective siRNA delivery and enhanced anti-tumor immunotherapy.

Blood-brain barrier-penetrating single CRISPR-Cas9 nanocapsules for effective and safe glioblastoma gene therapy

Targeting CD146 using folic acid-conjugated nanoparticles and suppression of tumor growth in a mouse glioma model
