Structural and binding studies of 2'- and 3-fucosyllactose and its complexes with norovirus capsid protein by molecular dynamics simulations

Summary: Human breast milk contains free oligosaccharides that play an important role in protecting breastfed infants from infectious diseases. Studying the structural and conformational models of these oligosaccharides is crucial for understanding their interaction with proteins. In this study, two trisaccharides in human breast milk were simulated and their conformational models were determined. The results showed that both trisaccharides exhibited high binding efficacy towards norovirus capsid protein, with 3-FL potentially being used as a preventive inhibitor for norovirus infection.

Engineering Escherichia coli for the High-Titer Biosynthesis of Lacto-N-tetraose

Summary: In this study, a high-titer LNT synthesis method was constructed in Escherichia coli. Through systematic design and metabolic engineering, a starting strain for LNT production was constructed by introducing lgtA and wbgO genes. The titer of LNT was further improved by overexpressing related genes. The final engineered strain produced a high titer of 31.56g/L of LNT in fed-batch cultivation.

Efficient Production of a Functional Human Milk Oligosaccharide 3'-Sialyllactose in Genetically Engineered Escherichia coli

Biosynthesis of Lacto-N-fucopentaose I in Escherichia coli by metabolic pathway rational design

The engineered strains produced high yields of LNFP I in fed-batch cultivation, with a conversion rate of lacto-N-tetraose to LNFP I reaching the highest level reported so far. The LNFP I-produced platform established here is broadly suitable for the production of complex and branched oligosaccharides.

The concentrations of HMOs in breast milk vary according to lactation period and mother's genetic status. Studies have shown significant variations in HMO concentrations for different types across lactation stages. In mature pooled human milk, the top 15 individual HMOs make up the majority of total HMO concentration.
