TREM-1 governs NLRP3 inflammasome activation of macrophages by firing up glycolysis in acute lung injury

Wen-Jing Zhong et al.

Summary: The activation of the triggering receptor expressed on myeloid cells-1 (TREM-1) can potentiate acute lung injury (ALI) and induce inflammation, but the underlying mechanism is unclear. This study found that blocking TREM-1 can attenuate the activation of NOD-, LRR- and pyrin domain-containing 3 (NLRP3) inflammasome and glycolysis in LPS-induced ALI mice. Furthermore, TREM-1 activation enhances glycolysis and inhibits oxidative phosphorylation in macrophages, and this effect is dependent on the hypoxia-inducible factor-1a (HIF-1 alpha)/mTOR/glycolysis pathway. Inhibiting mTOR or HIF-1 alpha can suppress TREM-1-induced metabolic reprogramming and NLRP3/caspase-1 activation. These findings reveal a novel mechanism of TREM-1-mediated NLRP3 inflammasome activation through the mTOR/HIF-1 alpha/glycolysis pathway.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Tumor cell-derived exosomes deliver TIE2 protein to macrophages to promote angiogenesis in cervical cancer

Shi Du et al.

Summary: In this study, it was found that the number of TEMs increased in cervical cancer tissues with high TIE2 expression and was associated with shorter survival. Further experiments revealed that exosomes derived from TIE2-high cervical cancer cells transported TIE2 protein directly to macrophages, leading to the induction of TEMs. These TEMs promoted tumor angiogenesis and exhibited an M2-like phenotype.

Interleukin-17 governs hypoxic adaptation of injured epithelium

Piotr Konieczny et al.

Summary: Mammalian cells activate HIFs to survive in low-oxygen environments, but injury-induced hypoxia is unable to activate HIF1 alpha. However, IL-17A derived from ROR gamma t(+) gamma delta T cells can activate HIF1 alpha. The IL-17A-HIF1 alpha axis promotes glycolysis for wound healing in epithelial cells, and loss of IL-17RC, HIF1 alpha, or blockade of glycolysis disrupts the repair process. These findings highlight the interconnectedness of inflammation, metabolism, and migration programs, as well as the role of immune cell-derived inputs in cellular adaptation to hypoxic stress during repair.

Toward personalized treatment approaches for non-small-cell lung cancer

Meina Wang et al.

Summary: Over the past two decades, molecular targeted therapies and immunotherapies have significantly improved outcomes for non-small-cell lung cancer (NSCLC), but the majority of advanced NSCLCs eventually become resistant. The future challenge lies in the application of predictive biomarkers for patient stratification in order to better manage NSCLC.
