Serine hydroxymethyltransferase 2 knockdown induces apoptosis in ccRCC by causing lysosomal membrane permeabilization via metabolic reprogramming

Summary: In this study, it was found that the expression of SHMT2 was upregulated in ccRCC tissues and associated with patient survival. Knockdown of SHMT2 inhibited the proliferation, migration, and invasion of ccRCC cells. Overexpression of SHMT2 promoted tumor progression. Mechanistically, SHMT2 depletion disrupted one-carbon metabolism, increased ROS levels, and decreased ATP levels through metabolic reprogramming, which disrupted cell homeostasis.

Anaplastic transformation in thyroid cancer revealed by single-cell transcriptomics

The transformation included stress-responsive, inflammatory, mitotic-defective, and mesenchymal ATC cells. We identified important milestones including diploid and aneuploid stages, with corresponding phenotypic changes and interactions with other cell types, providing new therapeutic opportunities for ATC treatment.

This study reveals that glycogen synthase kinase 3 (GSK3) plays a key role in regulating the expression of serine/one-carbon metabolic enzymes. Inhibition of nuclear GSK3 signaling enhances the efficacy of serine/one-carbon metabolism inhibitors in suppressing cancer cell proliferation.

Regulation and Therapeutic Targeting of MTHFD2 and EZH2 in KRAS-Mutated Human Pulmonary Adenocarcinoma

Summary: Activating KRAS mutations occur in about 30% of pulmonary adenocarcinoma cases, and specific inhibitors of G12C-mutated KRAS have improved the prognosis for about 14% of non-small cell lung cancer patients. This study shows the dependency of MTHFD2 and EZH2 expression on mutationally activated KRAS and their prognostic relevance in KRAS-mutated pulmonary adenocarcinoma, suggesting a potential therapeutic approach for NSCLC patients.
